      <description>&lt;p&gt;Several potential drivers of avian tooth loss have been proposed, although consensus remains elusive as fully toothless jaws arose independently numerous times among Mesozoic avialans and dinosaurs more broadly. The origin of crown bird edentulism has been discussed in terms of a broad-scale selective pressure or trend toward toothlessness, although this has never been quantitatively tested. Here, we find no evidence for models whereby iterative acquisitions of toothlessness among Mesozoic Avialae were driven by an overarching selective trend. Instead, our results support modularity among jaw regions underlying heterogeneous tooth loss patterns and indicate a substantially later transition to complete crown bird edentulism than previously hypothesized (∼90 mya). We show that patterns of avialan tooth loss adhere to Dollo's law and suggest that the exclusive survival of toothless birds to the present represents lineage-specific selective pressures, irreversibility of tooth loss, and the filter of the Cretaceous-Paleogene (K-Pg) mass extinction.Cell heterogeneity, such as antibiotic heteroresistance and cancer cell heterogeneity, has been increasingly observed. To probe the underlying molecular mechanisms in the dynamically changing heterogeneous cells, a high throughput platform is urgently needed to establish single cell genotype-phenotype correlations. Herein, we report a platform combining single-cell viability phenotypic analysis with digital molecular detection for bacterial cells. The platform utilizes polyethylene glycol hydrogel that cross-links through a thiol-Michael addition, which is biocompatible, fast, and spontaneous. To generate uniform nanoliter-sized hydrogel beads (Gelbeads), we developed a convenient and disposable device made of needles and microcentrifuge tubes. Gelbead-based single cell viability and molecular detection assays were established. Enhanced thermal stability and uncompromised efficiency were achieved for digital polymerase chain reaction (PCR) and digital loop-mediated isothermal amplification (LAMP) within the Gelbeads. Reagent exchange for in situ PCR following viability phenotypic analyses was demonstrated. The combined analyses may address the genotypic differences between cellular subpopulations exhibiting distinct phenotypes. The platform promises unique perspectives in mechanism elucidation of environment-evolution interaction that may be extended to other cell types for medical research.Femoroplasty is a proposed alternative therapeutic method for preventing osteoporotic hip fractures in the elderly. Previously developed navigation system for femoroplasty required the attachment of an external X-ray fiducial to the femur. We propose a fiducial-free 2D/3D registration pipeline using fluoroscopic images for robot-assisted femoroplasty. Intraoperative fluoroscopic images are taken from multiple views to perform registration of the femur and drilling/injection device. The proposed method was tested through comprehensive simulation and cadaveric studies. Performance was evaluated on the registration error of the femur and the drilling/injection device. In simulations, the proposed approach achieved a mean accuracy of 1.26±0.74 mm for the relative planned injection entry point; 0.63±0.21° and 0.17±0.19° for the femur injection path direction and device guide direction, respectively. In the cadaver studies, a mean error of 2.64 ± 1.10 mm was achieved between the planned entry point and the device guide tip.  Alert fatigue is a common issue with off-the-shelf clinical decision support. Most warnings for drug-drug interactions (DDIs) are overridden or ignored, likely because they lack relevance to the patient's clinical situation. Existing alerting systems for DDIs are often simplistic in nature or do not take the specific patient context into consideration, leading to overly sensitive alerts. The objective of this study is to develop, validate, and test DDI alert algorithms that take advantage of patient context available in electronic health records (EHRs) data. &lt;/p&gt;

&lt;p&gt;Data on the rate at which DDI alerts were triggered but for which no action was taken over a 3-month period (override rates) from a single tertiary care facility were used to identify DDIs that were considered a high-priority for contextualized alerting. A panel of DDI experts developed algorithms that incorporate drug and patient characteristics that affect the relevance of such warnings. The algorithms were then implemented as computable artifacts, validated using a synthetic health records data, and tested over retrospective data from a single urban hospital. &lt;/p&gt;

&lt;p&gt;Algorithms and computable knowledge artifacts were developed and validated for a total of 8 high priority DDIs. Testing on retrospective real-world data showed the potential for the algorithms to reduce alerts that interrupt clinician workflow by more than 50%. Two algorithms (citalopram/QT interval prolonging agents, and fluconazole/opioid) showed potential to filter nearly all interruptive alerts for these combinations. &lt;/p&gt;

Data commonly available in EHRs can improve DDI alert specificity.The recent development of liquid cell (scanning) transmission electron microscopy (LC-(S)TEM) has opened the unique possibility of studying the chemical behavior of nanomaterials down to the nanoscale in a liquid environment. Here, we show that the chemically induced etching of three different types of silica-based silica nanoparticles can be reliably studied at the single particle level using LC-(S)TEM with a negligible effect of the electron beam, and we demonstrate this method by successfully monitoring the formation of silica-based heterogeneous yolk-shell nanostructures. By scrutinizing the influence of electron beam irradiation, we show that the cumulative electron dose on the imaging area plays a crucial role in the observed damage and needs to be considered during experimental design.       <description>&lt;p&gt;We hope that in articulating these modeling assumptions and providing formal support for inclusive fitness maximization, we help bridge the gap between empiricists and theoreticians, which in some ways has been widening, demonstrating to mathematicians why biologists are content to use inclusive fitness, and offering one way to utilize inclusive fitness in general models of social behavior.Negative interspecific mating interactions, known as reproductive interference, can hamper species coexistence in a local patch and promote niche partitioning or geographical segregation of closely related species. Conspecific sperm precedence (CSP), which occurs when females that have mated with both conspecific and heterospecific males preferentially use conspecific sperm for fertilization, might contribute to species coexistence by mitigating the costs of interspecific mating and hybridization. We discussed whether two species exhibiting CSP can coexist in a local environment in the presence of reproductive interference. First, using a behaviorally explicit mathematical model, we demonstrated that two species characterized by negative mating interactions are unlikely to coexist because the costs of reproductive interference, such as loss of mating opportunity with conspecific partners, are inevitably incurred when individuals of both species are present. Second, we experimentally examined differences in mating activity and preference in two Harmonia ladybird species known to exhibit CSP. These behavioral differences may lead to local extinction of H. yedoensis because of reproductive interference by H. axyridis. This prediction is consistent with field observations that H. axyridis uses various food sources and habitats whereas H. yedoensis is confined to a less preferred prey item and a pine tree habitat. Finally, by a comparative approach, we observed that niche partitioning or parapatric distribution, but not sympatric coexistence in the same habitat, is maintained between species with CSP belonging to a wide range of taxa, including vertebrates and invertebrates living in aquatic or terrestrial environments. Taken together, it is possible that reproductive interference may destabilize local coexistence even in closely related species that exhibit CSP.Training in Population Ecology asks for scalable applications capable of embarking students on a trip from basic concepts to the projection of populations under the various effects of density dependence and stochasticity. Demography_Lab is an educational tool for teaching Population Ecology aspiring to cover such a wide range of objectives. The application uses stochastic models to evaluate the future of populations. Demography_Lab may accommodate a wide range of life cycles and can construct models for populations with and without an age or stage structure. Difference equations are used for unstructured populations and matrix models for structured populations. Both types of models operate in discrete time. Models can be very simple, constructed with very limited demographic information or parameter-rich, with a complex density-dependence structure and detailed effects of the different sources of stochasticity. Demography_Lab allows for deterministic projections, asymptotic analysis, the extraction of confidence intervals for demographic parameters, and stochastic projections. Stochastic population growth is evaluated using up to three sources of stochasticity environmental and demographic stochasticity and sampling error in obtaining the projection matrix.  The therapeutic use of psychedelics is regaining scientific momentum, but similarly psychoactive ethnobotanical substances have a long history of medical (and other) uses in indigenous contexts. Here we aimed to evaluate patient outcomes in a residential addiction treatment center that employs a novel combination of Western and traditional Amazonian methods. &lt;/p&gt;

&lt;p&gt;The study was observational, with repeated measures applied throughout treatment. All tests were administered in the center, which is located in Tarapoto, Peru. Data were collected between 2014 and 2015, and the study sample consisted of 36 male inpatients who were motivated to seek treatment and who entered into treatment voluntarily. Around 58% of the sample was from South America, 28% from Europe, and the remaining 14% from North America. click here We primarily employed repeated measures on a psychological test battery administered throughout treatment, measuring perceived stress, craving frequency, mental illness symptoms, spiritual well-being, and physical and emotional health. Addiction severity was measured on intake, and neuropsychological performance was assessed in a subsample from intake to at least 2 months into treatment. &lt;/p&gt;

&lt;p&gt;Statistically significant and clinically positive changes were found across all repeated measures. These changes appeared early in the treatment and were maintained over time. Significant improvements were also found for neuropsychological functioning. &lt;/p&gt;

However, in clinical practice, increasing numbers of patients present with raised PTH but normal serum calcium, renal function and vitamin D; this is known as normocalcaemic PHPT (nPHPT). Studies investigating the clinical presentation of this condition have shown that patients may present with hypertension, nephrolithiasis, impaired glucose tolerance, osteoporosis and fragility fractures. The prevalence of such complications in nPHPT is similar to that in classical hypercalcaemic PHPT (hPHPT). Although the National Institute for Health and Care Excellence (NICE) have developed guidelines for the management of PHPT generally, a consensus is yet to be reached on the optimal management of nPHPT specifically.       <description>&lt;p&gt;A series of heterocyclic compounds with a sulfonamide moiety were synthesized from reaction of enaminone 4 with active methylene compounds, glycine derivatives, 1,4-benzoquinone, hydroxylamine hydrochloride, hydrazonyl halides and dimethylacetylenedicarboxylate. The newly synthesized sulfonamide derivatives were characterized by FT-IR, 1H NMR, 13C NMR, mass spectroscopy, elemental analysis and alternative synthetic routes. The reactions products were evaluated for their antiproliferative activity against a panel of three different human cancerous cell lines, MCF-7 (breast), HepG-2 (liver) and HCT-116 (colon) and the results were deployed to derive the structure-activity relationships (SAR). Various test compounds were potent antiproliferative to cancerous cells; reaching very low micromolar levels, as in case of 21 which showed IC50 value of 6.2 μM against HepG-2 cell. In addition, treatment of cancerous cells with the synthesized compounds induced cell apoptosis and G2/M phase arrest evidenced by flow cytometric analysis. Furthermore, the activity of the synthesized compounds against TOP I and II were documented by DNA relaxation assays. Data revealed that compound 24 significantly interfered with TOP I- and II-mediated DNA relaxation, nicking and decatenation, with IC50 values 27.8 and 33.6 μM, respectively. Protein tyrosine phosphatase 1B (PTP1B) is emerging as a promising yet challenging target for drug discovery. To identify natural products as new prototypes for PTP1B inhibitors, we employed a hierarchical protocol combining ligand-based and structure-based approaches for virtual screening against natural product libraries. Twenty-six compounds were prioritized for enzymatic evaluation against PTP1B, and ten of them were recognized as potent PTP1B inhibitors with IC50 values at the micromolar level. Notably, nine compounds demonstrated evident selectivity to PTP1B over four other PTPs, including the most homologous T-cell protein tyrosine phosphatase (TCPTP). The results implicated that the structural uniqueness of the natural products might be a potential solution to the selectivity issue associated with the target PTP1B. The evolution of heat treatment for stone artefact production is a subject of major interest for our understanding of early modern humans. In this study, we examine the evidence from one region in Australia to provide a new record of the antiquity of heat treatment, explore chronological shifts in the frequency of heat treatment, and discuss the implications of these findings for early population dynamics and the technical knowledge early settlers might have brought with them. Until now, Australian heat treatment has only dated back 25000 years. This study of the Willandra Lakes, including Lake Mungo, has identified the oldest systematic evidence of heat treatment yet reported in Australia, dating to ∼42000 years. We also document time-dependent directional change in the frequency of the practice. At those early times, with over 60% of all silcrete artefacts heat-treated, we hypothesize that the practice was mastered and integrated as a recurrent technical solution to the complexities of knapping silcrete. Over time, the use of heat treatment decreased progressively until less than 10% of the artefacts were heat-treated in the terminal Holocene. This trajectory has implications for understanding the antiquity of heat treatment on the Australian continent and for investigating the factors that governed its use. Enamel thickness remains an important morphological character in hominin systematics and is regularly incorporated into dietary reconstructions in hominin species. We expand upon a previous study of enamel thickness in mandibular molars by examining a large maxillary molar sample of Plio-Pleistocene hominins (n = 62) and a comparative sample of extant nonhuman apes (n = 48) and modern humans (n = 29). 2D mesial planes of section were generated through microtomography, and standard dental tissue variables were measured to calculate average enamel thickness (AET) and relative enamel thickness (RET). AET was also examined across the lingual, occlusal, and buccal regions of the crown. This study confirms previous findings of increasing enamel thickness throughout the Plio-Pleistocene, being thinnest in Australopithecus anamensis and peaking in Australopithecus boisei, with early Homo specimens, exhibiting intermediate enamel thickness. Agreeing with previous findings, 2D plane of section enamel thickness is found to be a poor taxonomic discriminator, with no statistically significant differences observed between fossil hominins. For fossil hominins, modern humans, and Pongo, the occlusal region of enamel was the thickest, and the lingual enamel thickness was greater than buccal. Pan and Gorilla present the opposite pattern with enamel being thinnest occlusally. Comparison at each molar position between the maxilla and mandible revealed very few significant differences in fossil hominins but some evidence of significantly thicker maxillary enamel (AET) in modern humans and thinner maxillary enamel in Pan (RET). OBJECTIVE Pregnancies complicated by maternal preexisting diabetes have a 4-5-fold increased risk of stillbirth, and consequently routine antenatal nonstress testing (NST) was implemented into clinical practice decades ago. Though, international guidelines lack consensus and recommend anything from twice weekly testing from 32 weeks to once weekly testing from 38 weeks. The objective of this study was to examine how routine antenatal NST was used in centers with specific interest and dedication in the care of pregnant women with preexisting diabetes. STUDY DESIGN An electronic survey concerning the routine use of antenatal NST was sent to members of the European Diabetic Pregnancy Study Group (DPSG) between October 2016 and January 2017, representing in total 55 centers in 26 countries taking care of pregnant women with diabetes. RESULTS Answers from 38 centers (69.1 % (38/55)) in 22 countries were received. Based on real world information from these primarily European centers, anything from avoiding routine antenatal NST to testing twice weekly from early in third trimester in women with preexisting diabetes was reported.&lt;a href="https://www.selleckchem.com/products/Floxuridine.html" rel="noopener noreferrer"&gt;Floxuridine&lt;/a&gt;&lt;/p&gt;

      <description>&lt;p&gt;Efficiency of complex enrichment via acid-induced aggregation and subsequent centrifugation was assessed by yield and purity of protein in the pellet and pectin in the supernatant.Machine learning models are being utilized to provide wearable sensor-based exercise biofeedback to patients undertaking physical therapy. However, most systems are validated at a technical level using lab-based cross validation approaches. These results do not necessarily reflect the performance levels that patients and clinicians can expect in the real-world environment. This study aimed to conduct a thorough evaluation of an example wearable exercise biofeedback system from laboratory testing through to clinical validation in the target setting, illustrating the importance of context when validating such systems. Each of the various components of the system were evaluated independently, and then in combination as the system is designed to be deployed. The results show a reduction in overall system accuracy between lab-based cross validation (&amp;gt;94%), testing on healthy participants (n = 10) in the target setting (&amp;gt;75%), through to test data collected from the clinical cohort (n = 11) (&amp;gt;59%). This study illustrates that the reliance on lab-based validation approaches may be misleading key stakeholders in the inertial sensor-based exercise biofeedback sector, makes recommendations for clinicians, developers and researchers, and discusses factors that may influence system performance at each stage of evaluation.Origin recognition complex (ORC) binds to replication origins in eukaryotic DNAs and plays an important role in replication. Although yeast ORC is known to sequence-specifically bind to a replication origin, how human ORC recognizes a replication origin remains unknown. Previous genome-wide studies revealed that guanine (G)-rich sequences, potentially forming G-quadruplex (G4) structures, are present in most replication origins in human cells. We previously suggested that the region comprising residues 413-511 of human ORC subunit 1, hORC1413-511, binds preferentially to G-rich DNAs, which form a G4 structure in the absence of hORC1413-511. Here, we investigated the interaction of hORC1413-511 with various G-rich DNAs derived from human c-myc promoter and telomere regions. Fluorescence anisotropy revealed that hORC1413-511 binds preferentially to DNAs that have G4 structures over ones having double-stranded structures. Importantly, circular dichroism (CD) and nuclear magnetic resonance (NMR) showed that those G-rich DNAs retain the G4 structures even after binding with hORC1413-511. NMR chemical shift perturbation analyses revealed that the external G-tetrad planes of the G4 structures are the primary binding sites for hORC1413-511. The present study suggests that human ORC1 may recognize replication origins through the G4 structure.In this work, we present a battery-less wireless Micro-Electro-Mechanical (MEMS)-based respiration sensor capable of measuring the respiration profile of a human subject from up to 2 m distance from the transceiver unit for a mean excitation power of 80 µW and a measured SNR of 124.8 dB at 0.5 m measurement distance. The sensor with a footprint of ~10 cm2 is designed to be inexpensive, maximize user mobility, and cater to applications where disposability is desirable to minimize the sanitation burden. The sensing system is composed of a custom UHF RFID antenna, a low-loss piezoelectric MEMS resonator with two modes within the frequency range of interest, and a base transceiver unit. The difference in temperature and moisture content of inhaled and exhaled air modulates the resonance frequency of the MEMS resonator which in turn is used to monitor respiration. To detect changes in the resonance frequency of the MEMS devices, the sensor is excited by a pulsed sinusoidal signal received through an external antenna directly coupled to the device. The signal reflected from the device through the antenna is then analyzed via Fast Fourier Transform (FFT) to extract and monitor the resonance frequency of the resonator. By tracking the resonance frequency over time, the respiration profile of a patient is tracked. A compensation method for the removal of motion-induced artifacts and drift is proposed and implemented using the difference in the resonance frequency of two resonance modes of the same resonator.Streptococcus pneumoniae serotype 1 (ST1) was an important cause of invasive pneumococcal disease (IPD) globally before the introduction of pneumococcal conjugate vaccines (PCVs) containing ST1 antigen. The Pneumococcal Serotype Replacement and Distribution Estimation (PSERENADE) project gathered ST1 IPD surveillance data from sites globally and aimed to estimate PCV10/13 impact on ST1 IPD incidence. We estimated ST1 IPD incidence rate ratios (IRRs) comparing the pre-PCV10/13 period to each post-PCV10/13 year by site using a Bayesian multi-level, mixed-effects Poisson regression and all-site IRRs using a linear mixed-effects regression (N = 45 sites). After six years of PCV10/13 use, the all-site IRR was 0.05 (95% credibility interval 0.04-0.06) for all ages, 0.05 (0.04-0.05) for less then 5 years of age, 0.08 (0.06-0.09) for 5-17 years, 0.06 (0.05-0.08) for 18-49 years, 0.06 (0.05-0.07) for 50-64 years, and 0.05 (0.04-0.06) for ≥65 years. PCV10/13 use in infant immunization programs was followed by a 95% reduction in ST1 IPD in all ages after approximately 6 years. Limited data availability from the highest ST1 disease burden countries using a 3+0 schedule constrains generalizability and data from these settings are needed.The coronavirus disease 2019 (COVID-19) pandemic poses a worldwide healthcare challenge that needs an efficient response. Unfortunately, to date there is no highly effective treatment, so a deep understanding of COVID-19 risk factors could be an important step in treating the disease. Vitamin D affects the immune system in many different ways, and other authors already found that COVID-19 patients have low levels of vitamin D. In our retrospective study, we evaluated the vitamin D status at the time of hospital admission in 50 COVID-19 patients in Sicily, which is the southernmost region of Italy, and compared them with 100 control subjects matched for age and sex. Our data showed markedly low levels of vitamin D in patients with a positive polymerase chain reaction (PCR) for severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), but no association was found with inflammation markers or clinical severity.       <description>&lt;p&gt;Aqueous two-phase system (ATPS) is long seen as a technique that promotes higher purity and yield in a single step. It is witnessing increased acceptance as a preferred choice for combined goals of concentration, separation and purification of a target product, be it industrially valuable or environmentally contaminating. Purification of biomolecules like enzymes, proteins, nucleic acids, viruses, etc. has been the forte of ATPS. Currently, the technique is used for concentrating the toxic fractions from diverse industrial let offs, from food, dairy, beverage, pharmaceuticals, agriculture, dyeing, tannery and metal-processing industries. Apart from being simple, efficient, rapid, flexible, economical, and biocompatible, the selectivity, purity and yield are on par and sometimes higher than the traditional downstream steps. From an industrial angle too, problems related to scale-up of ATPS are being actively addressed. Many novel approaches are being added by way of varying ATPS components to increase the yield and purity. Another case in point is the inclusion of optimization techniques for zeroing in on the precise setting of the operating parameters. S1P Receptor antagonist With increasing impetus to the approach, we attempt to draw attention from academia and industries, alike, that are developing novel tweaks to the currently existing practices in ATPS. This review aims to assess and evaluate the different types of ATPS that have been used for the recovery of valuables and contaminants from industrial waste discharges.Environmental concerns due to fossil fuel usage has turned the research interest towards biomass and bioenergy field. Renewable biomass such as microalgae provides numerous advantages as they can grow in wastewater; sequester carbon dioxide, economical and eco-friendly. In this study, effect of pretreatment of microalgae (Scenedesmus obliquus) biomass using post-hydrothermal liquefaction wastewater (PHWW) for bio-oil production through hydrothermal liquefaction at a temperature of 300 °C was studied. Results showed liquefaction of pre-treated biomass yielded 48.53% bio-oil whereas 28.35% was resulted from biomass without pretreatment. The analysis of higher heating value of bio-oil showed that pretreated biomass oil has 36.19 MJ.Kg-1 against non-pretreated biomass oil, which has 28.88 MJ.Kg-1. Bio-oil (pretreated biomass) analysis revealed that 60% of compounds are in diesel and gasoline range with 58.09% of energy recovery. Bio-oil was rich in hydrocarbons of C7-C21 range with less oxygenated compounds. Carbon balance showed that an increase of 13% of carbon was sequestered in solid residue obtained from pretreated biomass and about 146% of increase also obtained in bio-oil.Studies have demonstrated that some commercial pet (i.e., cat and dog) food products contain high concentrations of mercury (Hg), and some products have Hg concentrations that are higher than expected based on the ingredients included in the package ingredient list. Additionally, concentrations of methylmercury, a particularly toxic form of Hg commonly associated with fish-based ingredients, are largely unstudied despite the widespread use of such ingredients in pet food products. This study aimed to quantify total Hg and methylmercury in a variety of commercial pet food products (n = 127), and use genetic tools to determine if specific ingredients contributed to high Hg concentrations in the final product. Results indicate that total Hg concentrations were above suggested maximum tolerable limits in three of the tested pet food products, and that methylmercury concentrations were at safe levels in all tested products. Next-generation amplicon sequencing using ten barcode primers was conducted to target distinct taxa and to determine if one primer set outperformed the others in amplifying the often heavily degraded DNA found in pet food products. The 16sUniF_16sUniR primer set generated a relatively higher number of reads across the broadest set of taxa, although several of the primer sets were useful in identifying common animal- and plant-based ingredients in commercial pet food products. Combined with the Hg results, it was demonstrated that pet food product ingredients are consistent among and between product lots. However, these results also revealed that adulteration is prevalent in pet food products.The huge excrement quantity from the increasing large-scale livestock stressed the ecological, environmental deterioration. As a major benefit for handling livestock manure, the slurry of biogas (BS) is developed during the production of biogas that might increase plant productivity. However, nitrous oxide (N2O) emissions from BS are considered a significant danger to the environment due to global warming potential. Furthermore, applying different proportions of BS combined with chemical fertilizer (CF) on N2O productions in the North China Plain (NCP) remains unclear. Herein, two sequential field trials were performed by maize-wheat rotations to substitute the CF by BS and reduce N2O emissions while keeping the crop yield stable. Four treatments were conducted, including T1, T3, T6, and CK. A total of 226.5 kg N ha-1 used in the maize-wheat rotation system. Additionally, different ratios of BS (100%, 80%, and 50%) combined with CF were used in wheat season in the tillering stage. Results showed integrated applications of BS with CF have potential for reducing N2O emission. Our findings showed that the maximum grain yield of CF was 6250 kg ha-1, which might be achieved by applying 38% BS and 62% of CF. This ratio yielded 1.03 kg ha-1 N2O emissions, which was 15% lesser than the N2O emission of CK, 1.21 kg ha-1. Considering whole growing period of wheat biogas treatments significantly reduced the cumulative N2O emissions from 17% to 26% compared to CF. To achieve maximum yield and minimum N2O emissions, an optimum 38% BS ratio has been suggested. The integrated use of BS and CF provided the greatest grain yield because of necessary nutrients provided by both slurry and CF. Consequently, N2O emissions reduced based on frequency and type of fertilizer. Physicochemical characterization of the nanoparticles was assessed using Transmission electron microscopy, Fourier transform infrared spectroscopy, Dynamic light scattering and X-ray diffraction technique. The swelling and in vitro drug release showed that the framework experiences pH-dependent swelling and release of Rifampicin. Rifampicin has lower release in acid medium and higher release in intestinal condition. Moreover, in view of the drug release results, the release kinetics and transport mechanisms were investigated and discussed. In vitro cytotoxicity assay demonstrated that the nanoparticles were non-toxic in nature. PIN1 proteins are a class of peptidyl prolyl cis-trans isomerases (PPIases), which have been implicated in numerous cellular functions like cell cycle progression, transcriptional control, signal transduction, promotion of oncogenesis and host-parasite interactions. In this work, the unfolding mechanism of a single domain PIN1 from Leishmania major (LmPIN1) has been characterized during thermal and denaturant-induced unfolding by differential scanning calorimetry (DSC), fluorescence and circular dichroism. Further, MD simulations have been performed to structurally probe the possible stages of its unfolding process. Both the fluorescence and CD data confirm classical two-state unfolding transitions for urea and GdnHCl. The thermal unfolding of LmPIN1, characterized by DSC, could optimally be fitted to a non two-state transition curve exhibiting two Tm's (53 °C and 57 °C) suggesting the possibility of an intermediate. Thermal unfolding of the modeled LmPIN1 by MD simulation shows that the unfolding process is initiated by increased fluctuations (dynamics) spanning residues 70-80, followed by perturbations in the sheet system and disjuncture of helix-sheet packing. Importantly, simulation and fluorescence quenching studies clearly suggest the possibility of the presence of residual structures of LmPIN1 even after complete denaturation. The goal of this work was to study the characteristics of a new phospholipid nanovesicular carrier for nasal administration of drugs. Multilamellar vesicles were visualized by electron microscopy, and their mean distribution size of 200 nm was evaluated by DLS. Measured pH and viscosity values were found adequate for a nasal delivery carrier. CLS micrographs of the nasal mucosa of rats following administration of the carrier incorporating probes with various properties show delivery into the nasal mucosa layers. Tramadol containing systems were characterized and tested for their analgesic effect in two pain animal models. In mice, a significantly higher antinociceptive effect and a rapid onset of action were obtained as compared to other nasal delivery carriers and to oral treatment. This enhanced analgesic effect was further confirmed in rat pain model and sustained by drug plasma and brain levels. To test the systems behavior in a larger animal, a pharmacokinetic crossover study was carried out in sheep after administrating Tramadol nasally in the nanocarrier and IV. The plasma and CSF absolute bioavailability values were 1.09 and 0.87, respectively. HPLC and LC-MS/MS methods for quantification of Tramadol in plasma, brain and CSF were developed and are presented here. It is noteworthy that no pathological alterations or inflammation signs were observed in rat nasal mucosa following sub-chronic treatment. The results obtained in this work encourage further investigation of using the new carrier for nasal delivery of drugs in humans. The physicochemical properties of camptothecin (CPT) limit its clinical application. To maximize drug efficacy, a novel intelligent prodrug delivery nanoplatform with a tumor microenvironment-cleavable core crosslinking strategy was proposed based on a phenylboronic acid (PBA) modified polyethylene glycol (PEG)-polyglutamic acid (PGlu) polymer with disulfide-bonded CPT, called PBA-PEG-P(Glu-co-GlussCPT). The fabricated nanoplatform was a spherical micelle that could withstand dilution and carry a large number of therapeutic molecules to the tumor tissues, thereby minimizing premature drug release. Moreover, the nanoplatform release 6.2 ± 0.62, 12.4 ± 1.8, 46.7 ± 0.33, and 79.2 ± 1.58% of CPT after incubation in 0.02, 1, 5, and 10 mM dithiothreitol for 24 h, respectively, exhibiting good reduction-sensitivity. Moreover, the nanoplatform exhibited significant antiproliferative activity against tumor cells. In addition, with PBA modification, the nanoplatform demonstrated enhanced endocytosis efficiency. This prodrug nanoplatform also exhibited significant in vivo antitumor efficacy on both murine and human hepatoma xenograft models, without showing significant systemic toxicity but demonstrating good biocompatibility. In other words, this novel intelligent prodrug delivery nanoplatform with tumor microenvironment-cleavable core crosslinking strategy and active targeting strategy based on prodrug polymer PBA-PEG-P(Glu-co-GlussCPT) demonstrated multiple functions and significant potential for antitumor drug delivery. The supercritical impregnation process was used as a green technology for the elaboration of drug delivery intraocular lenses to mitigate the risk of post-operatory endophthalmitis after cataract surgery. Commercially available hydrophobic acrylic (copolymer of benzyl methacrylate and methyl methacrylate) intraocular lenses (IOLs) were impregnated with gatifloxacin, a fourth generation fluoroquinolone drug, using pure supercritical CO2 (scCO2) to obtain solvent-free loaded implants. The interaction phenomena involved in the supercritical impregnation were studied by following in situ scCO2 sorption within the polymer support and the subsequent IOL swelling, and by taking into account drug solubility in the supercritical fluid phase. The drug impregnation yields determined though in-vitro release studies varied between 0.33 and 1.07 ± 0.07 μg·mg-1IOL in the studied experimental conditions (8 to 25 MPa, 308 to 328 K and 30 to 240 min impregnation duration).
