      <title>Ramifications involving SARS-CoV-2 infection for neurogastroenterology.</title>

      <description>&lt;p&gt;In advanced breast cancer (BCa) patients, not the primary tumor, but the development of distant metastases, which occur mainly in the organ bone, and their adverse health effects are responsible for high mortality. Targeted delivery of already known drugs which displayed potency, but rather unfavorable pharmacokinetic properties, might be a promising approach to overcome the current limitations of metastatic BCa therapy. Camptothecin (CPT) is a highly cytotoxic chemotherapeutic compound, yet poorly water-soluble and non-specific. Here, CPT was loaded into porous silicon nanoparticles (pSiNP) displaying the epidermal growth factor receptor (EGFR)-targeting antibody (Ab) cetuximab to generate a soluble and targeted nanoscale delivery vehicle for cancer treatment. After confirming the cytotoxic effect of targeted CPT-loaded pSiNP in vitro on MDA-MB-231BO cells, nanoparticles were studied in a humanized BCa bone metastasis mouse model. Humanized tissue-engineered bone constructs (hTEBCs) provided a humanized microenvironment for BCa bone metastases in female NOD-scid IL2Rgnull (NSG) mice. Actively targeted CPT-loaded pSiNP led to a reduction of orthotopic primary tumor growth, increased survival rate and significant decrease in hTEBC and murine lung, liver and bone metastases. This study demonstrates that targeted delivery via pSiNP is an effective approach to employ CPT and other potent anti-cancer compounds with poor pharmacokinetic profiles in cancer therapy. As the counterparts of noradrenaline and adrenaline in vertebrates, octopamine (OA) regulates multiple physiological and behavioral processes in invertebrate. OA mediates its effects via binding to specific octopamine receptors (OARs). Functional and pharmacological characterization of OARs have been reported in several insects. However, little work was documented in hemipteran insects. We cloned a β-adrenergic-like OAR (NcOA2B2) from Nephotettix cincticeps. NcOA2B2 shares high similarity with members of the OA2B2 receptor class. Transcript level of NcOA2B2 varied in various tissues and was highly expressed in the leg. After heterologous expression in CHO-K1 cells, NcOA2B2 was dose-dependently activated by OA (EC50 = 2.56 nM) and tyramine (TA) (EC50 = 149 nM). Besides putative octopaminergic agonists, dopaminergic agonists and amitraz and DPMF potently activated NcOA2B2 in a dose-dependent manner. Receptor activity was blocked by potential antagonists and was most efficiently antagonized by asenapine. Phentolamine showed both antagonist and agonist effects on NcOA2B2. Our results offer the important information about molecular and pharmacological characterization of an OAR from N. cincticeps that will provide the basis for forthcoming studies on its roles in physiological processes and behaviors, and facilitate the design of novel insecticides for pest control. The soybean aphid, Aphis glycines Matsumura (Hemiptera Aphididae) is a serious pest of the soybean plant, Glycine max, a major world-wide agricultural crop. We assembled a de novo genome sequence of Ap. glycines Biotype 1, from a culture established shortly after this species invaded North America. 20.4% of the Ap. glycines proteome is duplicated. These in-paralogs are enriched with Gene Ontology (GO) categories mostly related to apoptosis, a possible adaptation to plant chemistry and other environmental stressors. Approximately one-third of these genes show parallel duplication in other aphids. But Ap. gossypii, its closest related species, has the lowest number of these duplicated genes. An Illumina GoldenGate assay of 2,380 SNPs was used to determine the world-wide population structure of Ap. Glycines. China and South Korean aphids are the closest to those in North America. China is the likely origin of other Asian aphid populations. The most distantly related aphids to those in North America are from Austg natural history and host plant range. The impact of iron plaque (IP) on bioavailability of heavy metals to plants has been well documented, but the role of zinc (Zn) in modulating the associated processes remains elusive. We took Iris pseudacorus used in wetland for remediating Cd-contaminated water as an example and systematically studied the combined influence of Cd and Zn concentration on formation of IP and its consequence for immobilization and plant uptake of Cd. The experiment was conducted in hydroponic culture and in each treatment, we measured the physiological traits, activity of antioxidant enzymes (SOD, POD, CAT), mass of the IP, as well as the Cd content in both plant tissues and IP. The results showed that increasing Cd concentration resulted in a steady reduction in IP while the impact of zinc on IP was complicated and appeared to be coupled with Cd. When the Cd concentration was low (0.5 mg L-1 measured as CdCl2 2·5H2O) increasing Zn concentration reduced IP, while when the Cd concentration was increased to 5 mg L-1 increasing zi when the Zn concentration exceeded approximately 100 mg L-1 in all treatments, comparable with the change in immobilization and plant uptake of Cd as the Zn concentration increased. SEM analysis did prove the formation and variation of IP on the root surface of Iris pseudacorus in different treatments. We also found that the plant developed a survival strategy by scarifying its leaves with high Cd content. The results presented in this paper has wide implications as it revealed that care needs to be taken in applying Zn to enhance Cd immobilization and its plant uptake as exceeding the optimal application rate might reduce remediating efficiency rather than increase it. Agricultural production of Ligusticum chuanxiong Hort. is often affected by heavy metal pollution in soil, especially mixtures of cadmium (Cd) and lead (Pb). TGF-beta modulator We assessed metal-induced phytotoxicity in L. chuanxiong by exposing the plants to soil treated with Cd, Pb, or Cd/Pb mixtures. A combined Cd/Pb treatment alleviated the inhibition in plant growth, photosynthesis, and secondary metabolite generation seen in single-metal exposures in three of the four combinations. Most combined Cd/Pb treatments resulted in preferential uptake of magnesium, copper, and nitrogen in underground plant parts and accumulation of phosphorus and calcium in aboveground plant parts, thereby leading to improvements in photosynthetic potential. Compared with single-metal exposures, combined Cd/Pb treatment significantly decreased the contents of Cd by 16.67%-40.12% and Pb by 10.68%-21.70% in the plant, respectively.       <description>&lt;p&gt;We study the EDL force between two colloidal particles that are adsorbed to the surface of an electrolyte solution. The attachment of colloidal particles to a free surface of an electrolyte solution, which may interface with another liquid or vapor phase, is a well-known phenomenon that is employed in many scientific and industrial applications; the most well known of which is the Pickering emulsion. In addition to capillary stresses, the particles will experience an electrical double layer (EDL) force when they are close to each other. The force originates from the overlap of the diffusive layers of ions that appear in the electrolyte solution next to the charged surfaces of the particles and the charged surface of the electrolyte solution, which is free of particles. Here we elucidate the contribution of the free surface of the electrolyte solution to the EDL force between two spherical particles, which are half-submerged in the electrolyte solution. find more We solve the linearized Poisson-Boltzmann equation for the excess electrical potential near the particles and integrate over the resulting excess Maxwell and osmotic stresses on the particles. We further give corresponding P'ade approximations, thus enabling the use of simple formulae for the EDL force between interacting particles in cases similar to the ones in this study without the need to repeat the mathematical procedure employed here.Differences in the physical interaction between proteins, such as binding equilibria, can provide clues to differences in function. The binding of heat shock proteins to substrate proteins in living cells is such a case. Eukaryotic cells have evolved many homologs in the Hsp70 family of heat shock proteins, each specialized for a specific function. We previously showed that Hsp70, which is up-regulated during heat shock, binds the model substrate phosphoglycerate kinase (PGK) in human cells before PGK completely unfolds. We dubbed this the 'preemptive holding' mechanism. Here we study the homolog Hsc70 (heat shock cognate protein), which is constitutively expressed in human cells even in the absence of heat shock. Recent literature has demonstrated multiple functions performed by Hsc70 in cells under normal conditions. Despite the name 'heat shock cognate,' very few studies have shown whether Hsc70 is actually involved in heat shock response. Here we corroborate the existence of an in-cell heat shock response of Hsc70. We show that Hsc70 binds PGK in human cells in a cooperative manner that correlates directly with protein thermal unfolding. This 'unfolded state holding' mechanism differs from the Hsp70 'preemptive holding' mechanism. We rationalize the difference by protein evolution unlike Hsp70, which is upregulated to bind proteins specifically during heat shock, the finite amount of Hsc70 in cells cannot bind to still-folded proteins, or its multiple other functions would be compromised.Substitutional doping lanthanide ions (Ln3+) in CsPbX3 has been proven to be an efficient strategy for expanding the properties of the perovskite (PVK). Here, erbium (Er3+) uniformly doped CsPbX3 perovskite microplates are grown through a chemical vapor deposition method. Two fluorescence peaks at 430 and 520 nm which respectively correspond to the PVK and Er3+ emissions are observed. The time-resolved photoluminescence of both PVK host and Er dopants demonstrates that trap states play a critical role in facilitating the energy transfer between the PVK host and the Er dopants, which is vital to sensitizing the Er3+. A photophysical model was put forward to comprehensively describe this trap-mediated energy-transfer process, and the dynamics processes are modeled using correlated rate equations. The rates of the carrier's relaxation and energy transfer are respectively obtained as 6.6 and 49 ns-1, and a total energy transfer efficiency was obtained as ∼32.6%.Plasmonic nanostructures overcome Abbe's diffraction limit to create strong gradient electric fields, enabling efficient optical trapping of nanoparticles. However, it remains challenging to achieve stable trapping with low incident laser intensity. Here, we demonstrate Fano resonance-assisted plasmonic optical tweezers for single nanoparticle trapping in an array of asymmetrical split nanoapertures on a 50 nm gold thin film. A large normalized trap stiffness of 8.65 fN/nm/mW for 20 nm polystyrene particles at a near-resonance trapping wavelength of 930 nm was achieved. The trap stiffness on-resonance is enhanced by a factor of 63 compared to that of off-resonance due to the ultrasmall mode volume, enabling large near-field strengths and a cavity effect contribution. These results facilitate trapping with low incident laser intensity, thereby providing new options for studying transition paths of single molecules such as proteins.The fundamentals of using cracked film lithography (CFL) to fabricate metal grids for transparent contacts in solar cells were studied. The underlying physics of drying-induced cracks were well-predicted by an empirical correlation relating crack spacing to capillary pressure. CFL is primarily controlled by varying the crack template thickness, which establishes a three-way tradeoff between the areal density of cracks, crack width, and spacing between cracks, which in turn determine final grid transmittance, grid sheet resistance, and the semiconductor resistance for a given solar cell. Since CFL uses a lift-off process, an additional constraint is that the metal thickness must be less than 1/3 of the crack template thickness. The transmittance/grid sheet resistance/wire spacing tradeoffs measured in this work were used to calculate solar cell performance CFL-patterned grids should outperform screen-printed grids for narrow cells (0.5-2 cm wide) and/or cells with high semiconductor sheet resistance (≥100 Ω/sq), making CFL attractive for monolithically integrated thin-film photovoltaic modules.The accumulation of 99mTc-labeled probes targeting saturable systems of the body is hindered by the presence of a large excess of unlabeled ligands needed to ensure high radiochemical yields in a short reaction time.       <description>&lt;p&gt;OBJECTIVE To study reproducibility of f-wave parameters in terms of inter- and intrapatient variation. APPROACH Five parameters are investigated dominant atrial frequency (DAF), f-wave amplitude, phase dispersion, spectral organization, and spatiotemporal variability. For each parameter, the variance ratio R, defined as the ratio between inter- and intrapatient variance, is computed; a larger R corresponds to better stability and reproducibility. The study population consists of 20 high-quality ECGs recorded from patients with atrial fibrillation (11/9 paroxysmal/persistent). MAIN RESULTS The well-established parameters DAF and f-wave amplitude were associated with considerably larger R-values (13.1 and 21.0, respectively) than phase dispersion (2.4), spectral organization (2.4), and spatiotemporal variability (2.7). The use of an adaptive harmonic frequency tracker to estimate the DAF resulted in a larger R (13.1) than did block-based maximum likelihood estimation (6.3). SIGNIFICANCE This study demonstrates a noticeable difference in reproducibility among f-wave parameters, a result which should be taken into account when performing f-wave analysis. Light effects have been frequently used in volume rendering because they can depict the shapes of objects more realistically. Global illumination reflects light intensity values at relevant pixel positions of reconstructed images based on the considerations of scattering and extinction phenomena. However, in the cases of ultrasound volumes that do not use Cartesian coordinates, internal lighting operations generate errors owing to the distorted direction of light propagation, and thus increase the amount of light and its effects according to the position of the volume inside. selleck products In this study, we present a novel global illumination method with calibrated light along the progression direction in accordance with volume ray casting in non-Cartesian coordinates. In addition, we reduce the consumption of lighting operation in these lighting processes using a light-distribution template. Experimental results show the volume rendering outcomes in non-Cartesian coordinates that realistically visualize the global illumination effect. The light scattering effect is expressed uniformly in the top and bottom areas where many distortions are generated in the ultrasound coordinates by using the light template kernels adaptively. In this paper, we investigate the heating function of the nasal cavity qualitatively, using a high-quality, large-scale statistical shape model. This model consists of a symmetrical and an asymmetrical part and provides a new and unique way of examining changes in nasal heating function resulting from natural variations in nasal shape (as obtained from 100 clinical CT scans). Data collected from patients suffering from different nasal or sinus-related complaints are included. Parameterized models allow us to investigate the effect of continuous deviations in shape from the mean nasal cavity. This approach also enables us to avoid many of the compounded effects on flow and heat exchange, which one would encounter when comparing different patient-specific models. The effects of global size, size-related features, and turbinate size are investigated using the symmetrical shape model. The asymmetrical model is used to investigate different types of septal deviation using Mladina's classification. The qualitative results are discussed and compared with findings from the existing literature. Using electrospun fibers to deliver therapeutic agents has gained significant attention in various applications including cancer treatment and tissue regeneration. However, the effect of fluid flow and uptake by cells on the concentration profile is not well understood. In this study, we evaluated the release of lipophilic resveratrol from poly(ε-caprolactone) (PCL)-gelatin (GT) electrospun fibers experimentally and by using computational fluid dynamics (CFD). Resveratrol containing PCL-GT electrospun fibers were formed and used in a custom-built tubular bioreactor, to assess flow effect on concentration profile over 5 days. CFD model was developed to simulate release in both static cultures and under fluid flow conditions. Resveratrol stability in the culture medium and uptake by human umbilical vein endothelial cells and K562 cells over 3 days were used in the model. The concentration profile as a function of time was simulated and validated by experiments. The effects of inlet velocity, cellular uptake rate, bioreactor's length, and surrounding tissue porosity were assessed. The release profile was mainly affected by cellular uptake and the presence of porous media. The model suggests that the perfusion velocity might not have a significant effect relative to the cellular uptake rate and porosity of the surrounding tissue. Recognition and analysis of Diabetic Foot Ulcers (DFU) using computerized methods is an emerging research area with the evolution of image-based machine learning algorithms. Existing research using visual computerized methods mainly focuses on recognition, detection, and segmentation of the visual appearance of the DFU as well as tissue classification. According to DFU medical classification systems, the presence of infection (bacteria in the wound) and ischaemia (inadequate blood supply) has important clinical implications for DFU assessment, which are used to predict the risk of amputation. In this work, we propose a new dataset and computer vision techniques to identify the presence of infection and ischaemia in DFU. This is the first time a DFU dataset with ground truth labels of ischaemia and infection cases is introduced for research purposes. For the handcrafted machine learning approach, we propose a new feature descriptor, namely the Superpixel Colour Descriptor.       <description>&lt;p&gt;We report a case of 46-year-old male operated on for moderately differentiated lung adenocarcinoma. Postoperatively, he underwent six courses of chemotherapy and radiotherapy. He developed progressive severe inferior paraparesis accompanied by excruciating pain between the shoulders two years later. Magnetic resonance imaging revealed metastases in the bodies of T2 and T3 vertebrae with adjacent intradural extramedullary lesion compressing the spinal cord. The patient underwent surgical decompression and vertebral body cement augmentation that lead to pain relief and partial neurological recovery. The histological examination was consistent with metastases from low differentiated pulmonary adenocarcinoma. Surgical resection of intradural extramedullary metastasis improves patient quality of life by reducing pain intensity and neurological deficit. This is an open access article distributed under the terms of the CC0 Public Domain Dedication.Multimodal therapy, used for the treatment of patients with Hodgkin’s disease (HD), makes them prone to life-threatening infections, attributed mainly to febrile neutropenia. Herein, we present a case report of fatal combined bacterial and viral infection in a 49-year-old female patient, subject to polychemotherapy for HD. Rapid microbiological diagnosis performed by multiplex polymerase chain reaction elucidated the causes of the infection within hours. Listeria monocytogenes was detected in both the cerebrospinal fluid and blood samples. Nasopharyngeal swabs returned positive for two swine-derived strains of influenza A virus. We aimed to emphasize the importance of these pathogens and draw attention to their association in the aetiology of infections among patients receiving chemotherapy. In conclusion, better surveillance is needed to improve the early diagnosis of infectious complications in these patients.AIM To observe bacteremia following closed and open dental extraction. MATERIAL AND METHODS The study included two subject groups each comprising 29 participants. Group I patients received a single closed dental extraction, and group II patients – a single open extraction. Venous blood from the cubital vein of each patient was collected in three samples – preoperatively, 30 seconds after surgery, and 15 minutes after surgery. Anaerobic and aerobic hemocultures (Bact/ALERT, BioMerieux, Inc., Durhamn, N.C.) were used to study the bacteremia process. RESULTS Preoperatively, bacteremia was confirmed in 4 patients (6.9%) undergoing closed extraction and in 1 patient (1.7%) undergoing open extraction. Additionally, we found statistically significant relation between bacteremia and age (p=0.002). Thirty seconds after surgery bacteriemia was evident in 6 patients (10.3%) from group I and in 3 (5.2%) from group II. Fifteen minutes following surgery bacteremia was evident in 4 patients (6.9%) undergoing typical extraction and in one patient (1.7%) undergoing surgical extraction. No statistically significant relation between type of extraction and presence of bacteria in the bloodstream at 30 seconds (p=0.285) and at 15 minutes (p=0.166) was found. Coagulase-negative Staphylococcus was the most frequently found microorganism (22.2% of aerobic and 11.1% of anaerobic hemocultures). The results show greater significance of bacteremia at 30 seconds, compared to preoperative samples for both subject groups (p=0.03). CONCLUSIONS Bacteremia following dental extraction is unaffected by duration of intervention, type of extraction and gender. MATERIALS AND METHODS A cross-sectional study was performed in Plovdiv, October-December 2017 and January-March 2018 among 1826 school children aged 6-11. The studies have shown that effective drug must be able to inhibit more than one type of plasmepsin to avoid further growth of parasites and to prevent resistance of drug. Therefore, plasmepsins are believed to be excellent drug target for malarial disease. Extract of the plant Euphorbia hirta has been proved to exert antimalarial activity. However, molecular mechanism of this activity was not described. AIM The aim of present investigation is to identify antimalarial phytochemicals of Euphorbia hirta as plasmepsin protease inhibitors using an in silico approach. MATERIALS AND METHODS Docking studies were performed on three different protein targets plasmepsin I, II, and IV using iGEMDOCK. ADME and bioactivity predictions were done using molinspiration online tool. Toxicity studies were performed using ProTox-II online tool.
