      <title>Mitigation associated with urbanization effects upon water ecosystems through synchronous environmental restoration.</title>

      <description>&lt;p&gt;Although inhaled corticosteroids (ICSs) are the recommended first-line therapy for asthma, determining whether to continue or discontinue ICS treatment in patients with mild asthma remains challenging for clinicians. Several studies have revealed that patients with mild-persistent asthma maintained a well-controlled state after ICS withdrawal. However, the long-term outcomes of ICS withdrawal have not yet been determined. &lt;/p&gt;

&lt;p&gt;To determine the possible clinical outcomes of the discontinuation of ICS in patients with well-controlled mild asthma. &lt;/p&gt;

&lt;p&gt;We investigated the clinical outcomes of discontinuing ICSs in patients with well-controlled mild asthma and compared the time to loss of control (LOC) between patients who stopped ICS treatment (ICS withdrawal group, IWG) and those who continued treatment for 3 years (continuous ICS group, CIG). &lt;/p&gt;

&lt;p&gt;A significant difference in the time to LOC was observed between the IWG and CIG (hazard ratio, 2.56; 95% confidence interval, 1.52-4.33; P &amp;lt; .001). Increasing fractional exhaled nitric oxide levels (P = 0.008) and sputum eosinophil counts (%) (P = 0.015) revealed a weak but significant association with LOC risk in the CIG. The sputum eosinophil counts (P = 0.039) and serum total immunoglobulin E levels (P = 0.014) were significantly higher in the LOC group than in the non-LOC group of the CIG. &lt;/p&gt;

&lt;p&gt;Our results suggest that the maintenance of ICS treatment may help keep patients' asthma under control. Furthermore, patients with LOC had significantly higher sputum eosinophil counts in the CIG than those in the non-LOC group. Therefore, continuous ICS use by patients with mild, well-controlled asthma could be associated with good clinical outcomes. &lt;/p&gt;

 Separating individuals with viral-induced wheezing from those with asthma is challenging, and there are no guidelines for children under 6 years of age. Impulse oscillometry, however, is feasible in 4-year-old children. &lt;/p&gt;

&lt;p&gt;To explore the use of impulse oscillometry in diagnosing and monitoring asthma in young children and evaluating treatment response to inhaled corticosteroid (ICS). &lt;/p&gt;

&lt;p&gt;A total of 42 children (median age 5.3 years, range 4.0-7.9 years) with physician-diagnosed asthma and lability in oscillometry were followed for 6 months after initiation of ICS treatment. All children performed the 6-minute free-running test and impulse oscillometry at 3 time points. After the baseline, they attended a second visit when they had achieved good asthma control and a third visit approximately 60 days after the second visit. A positive ICS response was defined as having greater than 19 points in asthma control test and no hyperreactivity on the third visit. &lt;/p&gt;

&lt;p&gt;In total, 38 of 42 children responded to ICS treatment. Exercise-induced increases of resistance at 5 Hz decreased after ICS treatment (61% vs 18% vs 13.5%, P &amp;lt; .001), and running distance during the 6-minute test was lengthened (800 m vs 850 m vs 850 m, P=.001). Significant improvements in childhood asthma control scores occurred between the baseline and subsequent visits (21 vs 24 vs 24, P &amp;lt; .001) and acute physicians' visits for respiratory symptoms (1, (0-6) vs 0, (0-2), P=.001). Mutations in NOX subunit genes have been implicated in the pathogenesis of inflammatory bowel disease (IBD), indicating a crucial role for ROS in regulating host immune responses. In this study, we utilize genetically deficient mice to investigate whether defects in p40phox , one subunit of NOX, impair host immune response in the intestine and aggravate disease in an infection-based (Citrobacter rodentium) model of colitis. We show that p40phox deficiency does not increase susceptibility of mice to C. rodentium infection, as no differences in body weight loss, bacterial clearance, colonic pathology, cytokine production, or immune cell recruitment were observed between p40phox -/- and wild-type mice. Interestingly, higher IL-10 levels were observed in the supernatants of MLN cells and splenocytes isolated from infected p40phox -deficient mice. Further, a higher expression level of inducible nitric oxide synthase (iNOS) was also noted in mice lacking p40phox . In contrast to wild-type mice, p40phox -/- mice exhibited greater NO production after LPS or bacterial antigen re-stimulation.  Tumor-induced osteomalacia (TIO) is a paraneoplastic disorder, usually caused by benign mesenchymal tumors that produce high levels of the hormone fibroblast-growth-factor 23 (FGF23). The only curative therapy of the disease is resection of the causative tumors. This research was conducted to evaluate the efficacy of 18F-AlF-NOTA-octreotide ( 18F-OC) PET/CT in detecting TIO and its impact on patient management. &lt;/p&gt;

&lt;p&gt;Retrospective analysis of 17 patients with hypophosphatemic osteomalacia suspected of TIO was performed. 18F-OC PET/CT study was performed in all 17 patients to localize the tumor. 68Ga-DOTATATE PET/CT was performed in 4 out of 17 patients. 18F-OC and 68Ga-DOTATATE PET/CT studies were performed within 1 week of each other. Both studies were interpreted blindly without the knowledge of other imaging findings. The image findings were compared with the results of histopathological examinations and clinical follow-ups. &lt;/p&gt;

&lt;p&gt;18F-OC PET/CT scans were positive in 14 patients. Moreover, 4 out of 14 patients were performed with both 18F-OC and 68Ga-DOTATATE PET/CT. Both studies were able to localize the tumor in all the 4 patients. In total, 14 patients had surgery to remove the lesions. Postsurgical pathological examination confirmed causative tumors in these patients whose symptoms diminished promptly. The serum phosphate levels became normal confirming the diagnosis of TIO. 18F-OC PET/CT sensitivity, specificity and accuracy were 87.5%, 100% and 88.2% respectively. 18F-OC PET/CT findings affected patient management in 88.2% of cases. &lt;/p&gt;

Most yeast TF-based biosensors are built from natural promoters, resulting in large DNA parts retaining considerable homology to the host genome, which can complicate biological engineering efforts. There is a need to explore smaller, synthetic biosensors to expand the options for regulating gene expression in yeast. Here, we present a systematic approach to improving the design of an existing oxidative stress sensing biosensor in Saccharomyces cerevisiae based on the Yap1 transcription factor. Starting from a synthetic core promoter, we optimized the activity of a Yap1-dependent promoter through rational modification of a minimalist Yap1 upstream activating sequence. Our novel promoter achieves dynamic ranges of activation surpassing those of the previously engineered Yap1-dependent promoter, while reducing it to only 171 base pairs. We demonstrate that coupling the promoter to a positive-feedback-regulated TF further improves the biosensor by increasing its dynamic range of activation and reducing its limit of detection. We have illustrated the robustness and transferability of the biosensor by reproducing its activity in an unconventional probiotic yeast strain, Saccharomyces boulardii. Our findings can provide guidance in the general process of TF-based biosensor design.Environmental pH and gut transit rate are the key factors determining the dynamics of colonic microbiota. In this study, the effect of changing pH on the composition and metabolism of pooled faecal microbiota was elucidated at physiologically relevant dilution rates Dhigh = 0.2 and Dlow = 0.05 1/h. The results showed the best adaptability of Bacteroides ovatus within the pH range 6.0-8.0 at both dilution rates. The butyrate producing Faecalibacterium and Coprococcus comes were extremely sensitive to pH &amp;gt; 7.5, while the abundance of Akkermansia muciniphila increased significantly at pH &amp;gt;7 at Dhigh, causing a pH-dependant shift in the dynamics of mucin degrading species. Our study confirmed the importance of pH in the development of faecal microbiota in pectin-supplemented medium. Fermentation of other dietary fibres can be studied using the same approach.
