In recent decades, the regulatory role of long noncoding RNAs (lncRNAs) has been unmasked in glioma. In this study, we focused on the function and mechanism of LINC00641 in glioma. First of all, we found that LINC00641 was expressed at a low level in glioma cell lines. Importantly, overexpression of LINC00641 prevented cell proliferation but enhanced cell apoptosis. Meanwhile, NRGN, a previously-reported downregulated mRNA in GBM, was disclosed as a tumor suppressor in glioma cells. Besides, we verified that NRGN could be positively regulated by LINC00641 in glioma cells. Moreover, the cellular distribution of LINC00641 was identified to be cytoplasmic. Therefore, bioinformatics analysis and mechanism experiments were carried out and we determined that miR-4262 was the shared miRNA between LINC00641 and NRGN. In contrast to LINC00641 and NRGN, miR-4262 was dramatically upregulated in glioma cells. Furthermore, we confirmed that LINC00641 acted as a ceRNA in glioma cells via absorbing miR-4262 to upregulate NRGN. More importantly, silenced NRGN countervailed the repression on glioma cell proliferation caused by LINC00641 upregulation. Collectively, our findings unveiled that LINC00641 serves as a tumor inhibitor in glioma by targeting miR-4262/NRGN axis, providing a new potential therapeutic target for glioma patients.Despite literature highlighting numerous risks to the healthy psychosocial development of youth elite academy soccer players, little of this research is based on high-quality research designs. This study employed a prospective longitudinal cohort design to track psychosocial outcomes of academy involvement within male youth elite soccer players (n = 33, U12-U16 age groups) compared to age-matched soccer-active school pupils (n = 44) over 12 months. Participants completed questionnaires assessing the most commonly raised psychosocial concerns at four equally spaced data collection periods (T1-T4). Repeated measures multivariate analysis of variance (MANOVAs) indicated that, over the year, both groups reported a healthy and improving stress and recovery balance, as well as positive and stable needs satisfaction and physical, psychological and social well-being. selleckchem Academy players reported stable positive school-related quality of life, whereas school pupils reported increases from T3 to T4. We studied stress-induced cardiovascular, neuroendocrine, and behavioral responses of adult male F344 and LEW rats subjected to the single (120 min) or the repeated restraint stress (daily 120 min for 1 week). He was invited to the Polish-Lithuanian Commonwealth in the early 1760s and became head of the Faculty of Medicine at Vilnius University in 1781. In 1772, Bisius had authored the first original study on nervous and mental diseases in the Grand Duchy of Lithuania.       <description>&lt;p&gt;The pentasodium rare-earth tungstates Na5M(WO4)4 are closely related to the sodium rare-earth double tungstates Na5M(WO4)2 both adopting the scheelite structure type (space group I41/a, no. 88). After the preparation of polycrystalline powders via flux syntheses improving the phase purity significantly, the crystal structures of Na5M(WO4)4 (M = Y, La-Nd, Sm-Lu, Bi) were determined by single crystal XRD and Rietveld analysis. Na5M(WO4)4 is a promising phosphor material both as a host and as a 100% phosphor due to the possible charge transfer of the tungstate group and the absence of any concentration quenching. Na5M(WO4)4 incongruently melts to Na5M(WO4)2 and Na2WO4. After the clarification of the crystallographic relationship of Na5M(WO4)4 and Na5M(WO4)2 based on a rare isomorphic transition of index 5 (i5) the non-linear trend of the decomposition temperature within the row of rare earth ions is explained systematically taking into account the existence of domains within the crystal structure predetermining the posterior decomposition. A miscibility gap for solid solutions of Na5Y(WO4)4 and Na5Eu(WO4)4 or Na5Tb(WO4)4 is identified and its temperature dependence is investigated. Furthermore, the investigation of the fluorescent properties of Na5M(WO4)4 (M = Pr, Sm, Eu, Tb, Tm, Bi), Na5Y1-xEux(WO4)4 and Na5Y1-yTby(WO4)4 provided insights into the weak ligand field and the energy transfer from WO42- to M3+ governed by the emission of the sensitiser within Na5M(WO4)4. Additionally, the compounds were characterised by magnetic measurements and vibrational, UV/Vis and 151Eu Mössbauer spectroscopy.There is wide interest in developing efficient, robust and low-cost electrode materials for the electrolysis of water to produce clean hydrogen fuel. It is especially important to improve the performance and durability of electrocatalysts for the OER. Here we have shown that the transformation of nanoparticle (n-NNP) and crystalline (c-NNP) forms of mixed phosphate Na4Ni3(PO4)2P2O7 in highly alkaline solutions occurs along various routes and provokes the generation of 2D Ni(OH)2 nanosheets or stable core(phosphate)-shell(Ni(OH)2) particles, respectively. In both cases, in the carbon matrix (through chemical and electrochemical conversion of phosphate in situ during electrolysis in a 6 M KOH or NaOH solution) stable OER electrocatalysts with low overpotentials of 250-290 mV at a current density of 10 mA cm-2 were obtained. The best candidate for the OER process is core-shell particles, which maintain overpotentials of around 250 mV in 6 M KOH for more than 3 days. The activity enhancement can be attributed to the formation of abundant NiOOH nanoparticles on the shell surface due to improved lattice matching. This report discusses future prospects for the creation of core-shell particles to reduce the overpotential of durable electrocatalysts for the OER.A colorimetric sulfur dioxide (SO2) gas sensor based on a core-shell composite was developed. The composite was fabricated with a silicon dioxide core and a mesoporous MCM-41 shell (SiO2@MCM-41), and further loaded with a mixture of zinc chloride (ZnCl2), sodium nitroprusside (SNP) and hexamine as an SO2 indicator. The sensing properties of SiO2@MCM-41 toward SO2 were measured in solid powder, discs and a gas detection tube (GDT), respectively. Each of these sensing configurations showed a distinct color change from pale yellow to red, which indicates good potential for naked-eye detection of SO2. On the example of spherical nanoparticles consisting of a silicon core coated with sodium atoms, it is shown that the LSPR mode is selectively suppressed while the SPP mode is not. Such composite structures can be considered as an example of nanoplasmonic devices with selectively tuned characteristics.The development of novel multifunctional gene delivery systems with high efficiency is significant. Herein, due to the unique physical and optical properties of carbon dots (CDs), CDs prepared from polyethyleneimine (PEI) were modified with various hydrophobic chains and different degrees of substitution via an epoxide ring-opening reaction. The modification and substitution degree were confirmed using several analytical methods including 1H NMR spectroscopy, FT-IR spectroscopy, TEM, and XPS. These CDs were utilized as multifunctional, safe and efficient non-viral gene vectors. The results showed that these materials possessed capability for dual-channel imaging, which enabled the intracellular tracking of the delivered DNA. Both the type and substitution degree of the hydrophobic chain have a large influence on their transfection efficiency. Among the prepared CDs, Ole1.5-CD gave the highest transfection efficiency, which was up to 200 times higher than that of PEI 25 kDa in the presence of serum in A549 cells. Meanwhile, these CD materials showed much less cytotoxicity and better serum tolerance than the traditional cationic polymeric gene vector. The cellular uptake assay further confirmed the good serum tolerance and structure-activity relationship of the CD materials. The PXR-4βHC-LXR pathway could link the hepatic xenobiotic exposure and the regulation of cholesterol transport in peripheral tissues. Multinucleated osteoclasts formed through the fusion of mononuclear precursors are the principle cell capable of bone resorption. Pregnenolone (Preg) is the grand precursor of most if not all steroid hormones and have been suggested to be a novel anti-osteoporotic agent. However, the effects of Preg on osteoclast biology and function has yet to be shown. Here we examined the effect of Preg on receptor activator of nuclear factor kappa B ligand (RANKL)-induced osteoclast formation and bone resorption in vitro, and potential therapeutic application in inflammatory bone destruction and bone loss in vivo. Our in vitro cellular assays demonstrated that Preg can inhibit the formation of TRAP+ve osteoclast formation as well as mature osteoclast bone resorption in a dose-dependent manner. The expression of osteoclast marker genes CTSK, TRAP, DC-STAMP, ATP6V0d2, and NFATc1 were mai-osteoclastogenic and anti-resorptive agent for the potential treatment of osteolytic bone conditions. Molecular evidence indicates glioblastoma cells undergo an intriguing expansion of phenotypic properties to include neuron-like signaling using excitable membrane ion channels and synaptic proteins, augmenting survival and motility. Neurotransmitter receptors, membrane signaling, excitatory receptors, and Ca2+ responses are important candidates for the design of customized treatments for cancers within the heterogeneous central nervous system. Relatively few published studies of glioblastoma multiforme (GBM) have evaluated pharmacological agents targeted to signaling pathways in limiting cancer cell motility. Transcriptomic analyses here identified classes of ion channels, ionotropic receptors, and synaptic proteins that are enriched in human glioblastoma biopsy samples. The pattern of GBM-enriched gene expression points to a major role for glutamate signaling. However, the predominant role of AMPA receptors in fast excitatory signaling throughout the central nervous system raises a challenge on how to target inhibitors selectively to cancer cells while maintaining tolerability. This review critically evaluates a panel of ligand- and voltage-gated ion channels and synaptic proteins upregulated in GBM, and the evidence for their potential roles in the pathological disease progress. Evidence suggests combinations of therapies could be more effective than single agents alone. Natural plant products used in traditional medicines for the treatment of glioblastoma contain flavonoids, terpenoids, polyphenols, epigallocatechin gallate, quinones, and saponins, which might serendipitously include agents that modulate some classes of signaling compounds highlighted in this review. New therapeutic strategies are likely to exploit evidence-based combinations of selected agents, each at a low dose, to create new cancer cell-specific therapeutics. Danhong injection (DHI) is beneficial for ischemic stroke, but is prohibited for ICH due to risk of bleeding. The present study aims to explore the potential therapeutic time window and molecular mechanism of DHI in a collagenase-induced ICH model in aged rats. DHI administration after ICH could significantly improve body weight and neurological deficits, and reduce the hematoma volume and brain water content when compared to the vehicle control. Furthermore, the protective effect of DHI administration on days 1-3 after ICH was superior to those on days 3-5 or 7-9 after ICH. DHI remarkably increased the Peroxiredoxin 1 (Prx1) expression in astrocytes and reduced the expression of inflammatory factors tumor necrosis factor-α (TNF-α) and interleukin-β (IL-1β) after ICH. The immediate treatment of Prx1 inhibiter chelerythrine (Che) after ICH abolished the protective effect of DHI. Furthermore, the Che treatment reduced the expression of Prx1 in astrocytes, but increased the expression of TNF-α and IL-1β after ICH. DHI treatment could not reverse these changes. Therefore, the earlier DHI is administered, the better the neuroprotective effect. garsorasib DHI exerts antioxidative and anti-inflammatory function by increasing Prx1 in astrocytes. These present results may change the established understanding of DHI, and reveal a novel treatment approach for ICH. It has been used as a highly efficient compound for the treatment of cancer and hyperlipidemia, as well as possessing potential antiinflammatory properties. However, its effects on bone metabolism and osteoporosis haven't been reported. The purpose of our research were to reveal the biomolecular effects of asiatic acid on osteoclasts, and its underlying molecular mechanisms regulating its effects on receptor activator of NF-κB ligand (RANKL)-induced signaling pathways. We found that asiatic acid inhibited multinucleated tartrate-resistant acid phosphatase (TRAcP)-positive osteoclast differentiation and osteoclast induced bone loss. Real time PCR showed that asiatic acid reduced the expression of down-cascade target genes including Ctsk, Nfatc1, Calcr, and Atp6v0d2. Western blot and luciferase reporter gene assays revealed that asiatic acid inhibits RANKL mediated NF-κB and NFATc1 signalings. To determine whether these relationships differ depending on the sensorial phenotype. Methods Cross-sectional study performed on patients with type-2 diabetes-mellitus and neuropathy. Presence of DNP, pain intensity and phenotype, mood status, sleep characteristics and quality of life were measured. Results A total of 130 patients (65 with DNP) were included. DNP was related to poor sleep quality (OR = 1.03;CI95%1.02-1.05), pain treatment (OR = 3.00,CI95%1.24-7.29) or previous anxiety (OR = 2.70,CI95%1.05-6.99). Patients with specific phenotypes or depression (=0.82,CI95%-0.02-1.67) referred more severe pain. More complications were related to older age (OR = 1.40,CI95%1.12-1.66), higher pain intensity (OR = 1.51,CI95%1.00-2.28), lower cognitive performance (OR = 1.25,CI95%1.09-1.43), previous anxiety (OR = 10.48,CI95%1.46-75.24) and insulin treatment (OR = 124.50,CI95%6.64-2335.06). Decrease in mental quality of life was associated with sleep disorders (β = -0.33,CI95%-0.48,-0.23), physical comorbidities (β = -9.73,CI95%-18.15, -1.31) and previous anxiety (β = -7.91,CI95%-13.04, -2.77). Lower scores in physical quality of life were related to sleep disorders (β = -0.12,CI95%-0.21, -0.18), obesity (β = -8.35,CI95%-13.16, -3.55), longer time since diagnosis (β = -0.72,CI95%-1.44;0.01) and disability (β = -14.58,CI95%-24.69; -4.48). Conclusions The results support the idea that mental comorbidity and sleep disorders are factors associated with DNP and greater pain intensity, more diabetes complications and lower quality of life. Moreover, they highlight the relationship between sensorial phenotypes and pain intensity, and lower cognitive performance and diabetes complications.Objectives To evaluate the cost-effectiveness of screening children born at extremely low birth weight (ELBW) for hepatoblastoma using serial serum alpha-fetoprotein (AFP) measurements. Study design We created a decision tree to evaluate the cost-effectiveness of screening children born at ELBW between 3 months and 48 months of age compared with current standard of care (no screening). Our model used discounted lifetime costs and monetary benefits in 2018 U.S. dollars (USD), based on estimates in the published literature. The effects of uncertainty in model parameters were also assessed using univariate sensitivity analyses, in which we changed the values for one parameter at a time to assess the effect on the estimated incremental cost-effectiveness ratio (ICER). Results For the estimated 55,699 children born at ELBW in the United States each year, this screening is associated with 77.7 additional quality-adjusted life-years (QALYs) at a cost of $8.7 million. selleck This results in an ICER of about $112,000/QALY, which is considered cost-effective from a U.S. Societal perspective. For children diagnosed with hepatoblastoma, our model finds that the screening regimen is associated with 10.1% increase in survival, a 4.18 increase in expected QALYs, and a $245,184 decrease in expected cost. Conclusions Screening ELBW children for hepatoblastoma between 3 months and 48 months of age dominates the alternative and is cost-effective from a societal perspective.The social environment of animals can have profound implications on their behaviour and physiology. Naked mole-rats (Heterocephalus glaber) are highly social with complex dominance hierarchies that influence both stress- and reproduction-related hormones. Homeostasis may be affected by aggressive interactions, colony instability and social isolation. Furthermore, naked mole-rat colonies are characterised by a marked reproductive skew; a single female and few males are reproductively active while other colony members are reproductively suppressed. Thus, there are distinct differences in related hormone concentrations between reproductively active and non-active animals; however, this changes when non-reproductive individuals are removed from the colony. We investigated the effects of social isolation and colony disruption on plasma cortisol and progesterone concentrations in non-breeding naked mole-rats. During colony disruption, we found a significant increase in cortisol concentrations in females removed frothe colony structure may affect the sensitivity of the animals to changes in their environment.Wild animals are brought into captivity for many reasons. However, unlike laboratory-bred animals, wild caught animals often respond to the dramatic shift in their environment with physiological changes in the stress and reproductive pathways. Using wild-caught male and female house sparrows (Passer domesticus) we examined how time in captivity affects the expression of reproductive and stress-associated genes in the brain, specifically, the hypothalamus. We quantified relative mRNA expression of a neurohormone involved in the stress response (corticotropin releasing hormone [CRH]), a hypothalamic inhibitor of reproduction (gonadotropin inhibitory hormone [GnIH]), and the glucocorticoid receptor (GR), which is important in terminating the stress response. To understand potential shifts at the cellular level, we also examined the presence of hypothalamic GnIH (GnIH-ir) using immunohistochemistry.       <description>&lt;p&gt;Coarsening dynamics in both LRO and QLRO regimes are further characterized by power-law growth with disorder-dependent exponents within our simulation timescales. The growth exponents in the LRO regime decrease from 0.5 in the pure case to 0.22 in the maximum disordered case, whereas the corresponding change in the QLRO regime happens from 0.45 to 0.38. We further explored the coarsening dynamics in the bond-diluted clock model and, in both the models, the effect of the disorder is more significant for the quench in the LRO regime compared to the QLRO regime.The equilibrium properties of a finite-length linear polymer chain confined in an infinite wedge composed of two perfectly reflecting hard walls meeting at a variable apex angle (α) are presented. One end of the polymer is anchored a distance y from the apex on the conical axis of symmetry, while the other end is free. We report here, the nonmonotonic behavior of θ temperature as a function of y for a finite-length chain. Data collapse for different chain lengths indicates that such behavior will exist for all finite lengths. We delineate the origin of such nonmonotonic behavior, which may have potential applications in understanding the cellular process occurring in nanoconfined geometries.Colloidal particles, which are ubiquitous, have become ideal testing grounds for the structural glass transition theories. In these systems glassy behavior arises as the density of the particles is increased. Thus, soft colloidal particles with varying degree of softness capture diverse glass-forming properties, observed normally in molecular glasses. Brownian dynamics simulations for a binary mixture of micron-sized charged colloidal suspensions show that tuning the softness of the interaction potential, achievable by changing the monovalent salt concentration results in a continuous transition from fragile to strong behavior. Remarkably, this is found in a system where the well characterized interaction potential between the colloidal particles is isotropic. buy MEDICA16 We also show that the predictions of the random first-order transition (RFOT) theory quantitatively describes the universal features such as the growing correlation length, ξ∼(ϕ_K/ϕ-1)^-ν with ν=2/3 where ϕ_K, the analog of the Kauzmann temperature, depends on the salt concentration. As anticipated by the RFOT predictions, we establish a causal relationship between the growing correlation length and a steep increase in the relaxation time and dynamic heterogeneity as the system is compressed. The broad range of fragility observed in Wigner glasses is used to draw analogies with molecular and polymer glasses. The large variations in the fragility are normally found only when the temperature dependence of the viscosity is examined for a large class of diverse glass-forming materials. In sharp contrast, this is vividly illustrated in a single system that can be experimentally probed. Our work also shows that the RFOT predictions are accurate in describing the dynamics over the entire density range, regardless of the fragility of the glasses.Different attempts to describe financial markets, and stock prices in particular, with the tools of statistical mechanics can be found in the literature, although a general framework has not been achieved yet. In this paper we use the physics of many-particle systems and the typical concepts of soft matter to study two sets of US and European stocks, comprising the biggest and most stable companies in terms of stock price and trading. Upon correcting for the center-of-mass motion, the structure and dynamics of the systems are studied (in the European set, the structure is studied for the UK subset only). The pair distribution of the stocks, corrected to account for the nonuniform distribution of prices, is close to 1, indicating that there is no direct interaction between stocks, similar to an ideal gas of particles. The dynamics is studied with the mean-squared price displacement (MSPD); the price correlation function, equivalent to the intermediate scattering function; the price fluctuation distribution; anoled systems.We report experimental studies on the phase behavior of binary mixtures of 1″,7″-bis(4-cyanobiphenyl-4'-yl)heptane (CB7CB) and 4,4-diheptyloxyazoxybenzene, which exhibit, apart from the nematic (N) and twist-bend nematic (N_TB) phases, the induced smectic-A (Sm-A) phase for weight fraction of CB7CB between 0.05 and 0.70. In planar nematic layers, the N_TB phase separates as droplets of tactoidlike planform; the chirality of droplets manifests in the optical dissimilarity between their opposite angular ends. Our main result is that, in the appropriate two phase region, Sm-A nuclei with positive dielectric anisotropy change over to disks immersed in the nematic above some electric field, their edges decorated by periodic bright spots, a result which was earlier reported in another binary system exhibiting the induced Sm-A phase [R. Pratibha and N. V. Madhusudana, Physica A 224, 9 (1996)10.1016/0378-4371(95)00311-8]. We develop a simple theory for the threshold of this distortion, which is a periodic undulation of the edge of the disk, demonstrating that it arises from saddle-splay elasticity of Sm-A, the low Sm-A-N interfacial tension unable to suppress the distortion. The observed increases in the number of bright spots with field, and with the radius of the disk at a given field, in both the experimental systems are also accounted for by the model. The distortion, which results in the most direct visualization of saddle splay in Sm-A, is also exhibited by disks nucleating on surfaces treated for homeotropic anchoring.We present direct measurements of electron temperature variations within an inertially confined deuterium-tritium plasma caused by localized mix of higher-Z materials into the central hot spot. The data are derived from newly developed differentially filtered penumbral imaging of the bremsstrahlung continuum emission. Our analysis reveals distinct localized emitting features in the stagnated hot-spot plasma, and we infer spatial variations in the electron temperature the mixed region is 660±130eV colder than the surrounding hot-spot plasma at 3.26±0.11keV.
