However, the mechanisms by which plants sense temperature are not well understood. The evening complex is a major signalling hub and a core component of the plant circadian clock2,3. The evening complex acts as a temperature-responsive transcriptional repressor, providing rhythmicity and temperature responsiveness to growth through unknown mechanisms2,4-6. The evening complex consists of EARLY FLOWERING 3 (ELF3)4,7, a large scaffold protein and key component of temperature sensing; ELF4, a small α-helical protein; and LUX ARRYTHMO (LUX), a DNA-binding protein required to recruit the evening complex to transcriptional targets. ELF3 contains a polyglutamine (polyQ) repeat8-10, embedded within a predicted prion domain (PrD). Here we find that the length of the polyQ repeat correlates with thermal responsiveness. We show that ELF3 proteins in plants from hotter climates, with no detectable PrD, are active at high temperatures, and lack thermal responsiveness. The temperature sensitivity of ELF3 is also modulated by the levels of ELF4, indicating that ELF4 can stabilize the function of ELF3. In both Arabidopsis and a heterologous system, ELF3 fused with green fluorescent protein forms speckles within minutes in response to higher temperatures, in a PrD-dependent manner. A purified fragment encompassing the ELF3 PrD reversibly forms liquid droplets in response to increasing temperatures in vitro, indicating that these properties reflect a direct biophysical response conferred by the PrD. To facilitate an agile response to the pandemic, we developed How We Feel, a web and mobile application that collects longitudinal self-reported survey responses on health, behaviour and demographics. Here, we report results from over 500,000 users in the United States from 2 April 2020 to 12 May 2020. We show that self-reported surveys can be used to build predictive models to identify likely COVID-19-positive individuals. We find evidence among our users for asymptomatic or presymptomatic presentation; show a variety of exposure, occupational and demographic risk factors for COVID-19 beyond symptoms; reveal factors for which users have been SARS-CoV-2 PCR tested; and highlight the temporal dynamics of symptoms and self-isolation behaviour. These results highlight the utility of collecting a diverse set of symptomatic, demographic, exposure and behavioural self-reported data to fight the COVID-19 pandemic.Inflammatory bowel disease (IBD) is a complex disorder that imposes a growing health burden. Multiple genetic associations have been identified in IBD, but the mechanisms underlying many of these associations are poorly understood. Animal models are needed to bridge this gap, but conventional laboratory mouse strains lack the genetic diversity of human populations. To more accurately model human genetic diversity, we utilized a panel of chromosome (Chr) substitution strains, carrying chromosomes from the wild-derived and genetically divergent PWD/PhJ (PWD) strain on the commonly used C57BL/6J (B6) background, as well as their parental B6 and PWD strains. Two models of IBD were used, TNBS- and DSS-induced colitis. Compared with B6 mice, PWD mice were highly susceptible to TNBS-induced colitis, but resistant to DSS-induced colitis. Using consomic mice, we identified several PWD-derived loci that exhibited profound effects on IBD susceptibility. The most pronounced of these were loci on Chr1 and Chr2, which yielded high susceptibility in both IBD models, each acting at distinct phases of the disease. Leveraging transcriptomic data from B6 and PWD immune cells, together with a machine learning approach incorporating human IBD genetic associations, we identified lead candidate genes, including Itga4, Pip4k2a, Lcn10, Lgmn, and Gpr65.Technologies that rely on quantum bits (qubits) require long coherence times and high-fidelity operations1. The effect of ionizing radiation leads to an elevated quasiparticle density, which we predict would ultimately limit the coherence times of superconducting qubits of the type measured here to milliseconds. We further demonstrate that radiation shielding reduces the flux of ionizing radiation and thereby increases the energy-relaxation time. The films did not exhibit antimicrobial activity against bacteria or yeasts.Improved ocular delivery of a poorly soluble anti-glaucoma drug, acetazolamide (ACZ), in a stable nanosuspension (NS) was the main target of the study. The anionic polypeptide, poly-γ-glutamic acid (PG) and the glycosaminoglycan, hyaluronic acid, were used to stabilize ACZ-NS prepared using the antisolvent precipitation (AS-PT) coupled with sonication technique. The synthesized CS/ZnO nanocomposite was characterized by using UV-visible spectroscopy, X-ray diffraction (XRD), field emission-scanning electron microscopy (FE-SEM), transmission electron microscopy (TEM), energy dispersive X-ray spectroscopy (EDS), fourier transform infrared spectroscopy (FTIR) techniques. Electron microscopy images showed agglomeration of CS/ZnO nanocomposite having spherical shaped structure with an average size of 21-47 nm. The observed bands around 400-500 cm-1 in the IR spectrum indicated the presence of metal‑oxygen bond, whereas bands at 1512 and 1745 cm-1 indicated the presence of amine groups (-NH2) which confirms the presence of CS in the CS/ZnO nanocomposite.       <description>&lt;p&gt;For different size classes, significant clumped distributions were observed for the regeneration levels (1-3), while the spatial distribution of tree layers (4-5) usually fluctuated distinctly among various distribution patterns. The scale effects of different size classes were mainly dominated by the linear increases (44%), the power increases (15%) and the negative quadratic polynomials (12%). For each forest type and sampling scale, the cluster degrees of trees decreased significantly with increasing tree sizes. Within each forest type, the pattern size of non-dominant species was significantly larger than that of dominant species, while the pattern size of regeneration layers was significantly larger than that of tree layers.To explore the effects of different coated slow-release calcium peroxide on soil microbial characteristics in gleyed paddy field, we set seven treatments by simulation test in a greenhouse, including CK (none calcium peroxide), calcium peroxide powder, calcium peroxide particle and other four coated slow-release calcium peroxide treatments. Samples were collected in same tillering stage of early season rice. The results showed that all the applications of calcium peroxide could improve the concentrations of active soil organic carbon, soil available nutrients, soil microbial biomass, culturable microorganism population, as well as soil enzyme activity. The coated slow-release calcium peroxide had stronger effects than calcium peroxide particles and calcium peroxide powder. The calcium peroxide coated by ethyl cellulose was the most effective, which enhanced soil active organic carbon, soil microbial biomass carbon, nitrogen, phosphorus by 19.4%, 11.4%, 121.5% and 127.2%, soil alkaline hydrolysis nitrogen and available phosphorus by 4.0% and 45.5%, soil culturable bacteria and culturable microorganism population by 137.3% and 113.7%, fungi and actinomyces number were increased by 33.6% and 44.7%. The enzyme activities of invertase, phosphatase, urease as well as catalase were increased by 92.4%, 91.8%, 112.5% and 17.1%, respectively. The results could provide reference for improving gleyed paddy field with coated calcium peroxide.Long-term fertilization can change the supply of soil carbon and nitrogen (N), with consequences on the abundance and community structure of soil microorganisms. Based on the long-term fertilization positioning experiment station of brown earth, we analyzed the dynamics of soil ammonia-oxidizing archaea (AOA) and ammonia-oxidizing bacteria (AOB) under different fertilization treatments, including no fertilization (CK), low level of inorganic N fertilizer (N2), high level of inorganic N fertilizer (N4), and organic manure combined with inorganic N fertilizer (M2N2), aiming to provide a basis for microbiological mechanism of soil N transformation and improvement of soil fertility. The results showed that the ratio of AOA to AOB abundance was 2.28-61.95 under different fertilization treatments. NIBRLTSi Compared with that in CK, the AOA abundance was reduced by 1.6%-13.6% after long-term fertilization. The abundance of AOB in N4 treatment decreased first and then increased with soil depths, but with contrary results in other treatments. The Shannon diversity index (H), evenness index (J), and Simpson index (S) of AOB were higher than those of AOA. The AOB diversity was increased at 0-20 cm soil layer in M2N2 treatment, while that of AOA was decreased. Soil AOB clustered with soil depths, and neither AOA nor AOB community clustered with fertilization treatments. In summary, long-term fertilization altered the composition of AOA and AOB. AOA was sensitive to environment, whereas AOB was more abundant and stable.Soil salinization severely hinders the development of agricultural economy in the Yellow River Delta. Clarifying the spatial variability of soil salinity at multiple scales in the field is of great significance for the improvement and utilization of saline soils and agricultural production. In this study, by dividing the three dimensions of field, plot and ridge, we collceted 152 sets of conducti-vity data through field survey sampling in a summer maize field in Kenli County of the Yellow River delta. The methods of classic statistics, geostatistics and Kriging interpolation were used to analyze the spatial variability and scale effects of multi-scale soil salt in the field. The results showed that soil in this area was moderately salinized, with the extent of soil salinity moderately varying at three scales. From the field, plot to the ridge scale, with the decreases of sampling scale, the variability of soil salinity increased and the standard deviation increased. The ridge and plot scales showed strong spatial correlation. The optimal model was Gaussian model, which was mainly affected by structural factors. The field scale was of medium spatial correlation, with exponential model as the optimal one, which was influenced by both random factors and structural factors. The spatial distribution characteristics of soil salinity at different scales were significantly different. The spatial chara-cteristics at small scale were masked at large scale, showing obvious scale effect. The distribution of soil salinity at the micro-ridge scale between ridges had obvious variation. Soil salt content gradually decreased with the micro-topography from high to low, while vegetation coverage changed from sparse to dense.Effects of fermented apple products on the growth of continuous cropping Malus hupehensis Rehd. seedlings and soil environment were examined in a pot experiment to provide theoretical basis for apple replant disease. T2 and T3 increased the activities of nutrient conversion-related enzymes in continuous cropping soil.       <description>&lt;p&gt;The main purpose of current study is accurate prediction of NOx emissions from diesel engines considering in-cylinder ion current. To reach this goal, a validated thermodynamic multi-zone model was used. A modified chemical kinetics mechanism of diesel fuel oxidation was used too. A chemical kinetics mechanism of NOX formation including 103 reactions was added to the main mechanism. A set of ions and ionic reactions was added to the developed chemical kinetics mechanism and finally a modified chemical kinetics mechanism with 445 reactions and 100 species was formed. The developed mechanism was coupled to the multi-zone model and a diesel engine was simulated. The importance of Zeldovich mechanism, prompt mechanism, N2O mechanism and NNH mechanism were investigated. The progress rates of reactions were calculated and important reactions were identified. The results show that the oxygenated ions, NO+, O+ and O2+, has more effects on NO production than other ions. The prompt mechanism plays an important role in predicting the ion current inside the chamber. Because this mechanism has reactions that can lead to CH production. The CH radicals produced by this mechanism can be employed by basic ionic reactions and lead to ion production. The results show that using NOx related ionic reactions results in accurate prediction of engine exhaust NOx.Sediments serve as both source and sink of contaminants (e.g., Cu) and biologically important materials (e.g., metals, nutrients). Bioturbation by benthic organisms is ecologically relevant as bioturbation affects the physio-chemical characteristics of sediments, thus altering nutrient and contaminant distribution and bioavailability. We examined the effects of sediment-associated Cu on T. tubifex with conventional toxicity endpoints, such as mortality and growth, and less commonly used non-destructive endpoints, such as bioturbation and feeding. An experimental approach was developed to examine the applicability of simple methods to detect effects on bioturbation and feeding. Two experiments were conducted with 7-day exposures to uncontaminated or Cu-spiked natural sediment at six Cu concentrations to examine Cu bioaccumulation and effects. Endpoints included worm mortality, feeding rate and growth (experiment A) and worm bioturbation (particle diffusion and maximum penetration depth, experiment B). A microparticle tracer was placed on the sediment surface and vertical particle transport was followed over time. Adverse effects were detected for all endpoints (bioturbation, feeding rate, growth and survival) a slight positive effect at the lowest Cu concentrations followed by adverse effects at higher concentrations indicating hormesis. These simple, non-destructive endpoints, provided valuable information and demonstrated that sediment-associated contaminants, such as Cu, can influence bioturbation activity, which in turn may affect the distribution of sediment-bound or particulate pollutants, such as the plastic microparticles studied here. Thus, we suggest to use simple endpoints, such as bioturbation and feeding rate, in ecotoxicity testing since these endpoint account for the influence of interactions between pollutants and benthos and, thus, increase ecological relevance.The development of photo-polymerizable ferrous sulfate liposomes has been perused for iron fortification of food. Iron fortification is accompanied by several limitations that reduce its quality as it provokes sensory problems due to the oxidation of Fe2+ into Fe3+. To overcome such undesirable effects and improve the bioavailability of iron, photo-polymerizable 1,2-bis(10,12-tricosadiynoyl)-sn-glycero-3-phosphocholine (DC8,9PC) phospholipids have been employed as iron delivery system. DC8,9PC possesses diacetylene groups that undergo intramolecular cross-linking following UV treatment, resulting in enhanced stability, high encapsulation efficiency (~91%) without inducing sensory changes during milk fortification, along with less oxidation and con-tent leakage after long term storage in ethanol. During the trial, hematological parameters, renal and hepatic toxicity were evaluated. Serum urea, creatinine, alkaline phosphatase, transaminases (GOT and GPT), as well as hemogram results, were evaluated before the first administration and during the second and third weeks after the start of the treatment. Whether beneficial effects of LY2828360 are dependent upon the presence of a pathological pain state are unknown and its impact on unwanted opioid-induced side-effects have never been investigated. Here, we asked whether LY2828360 would produce synergistic anti-allodynic effects with morphine in a paclitaxel model of chemotherapy-induced neuropathic pain and characterized its impact on opioid-induced reward and other unwanted side-effects associated with chronic opioid administration. Isobolographic analysis revealed that combinations of LY2828360 and morphine produced synergistic anti-allodynic effects in suppressing paclitaxel-induced mechanical allodynia. In wildtype (WT) mice, LY2828360 blocked morphine-induced reward in a conditioned place preference assay without producing reward or aversion when administered alone. The LY2828360-induced attenuation of morphine-induced reward was absent in CB2 knockout (CB2KO) mice. In the absence of a neuropathic pain state, LY2828360 partially attenuated naloxone-precipitated opioid withdrawal in morphine-dependent WT mice, and this withdrawal was itself markedly exacerbated in CB2KO mice. Moreover, LY2828360 did not reliably alter morphine-induced slowing of colonic transit or attenuate tolerance to morphine antinociceptive efficacy in the hot plate test of acute nociception. Our results suggest that cannabinoid CB2 receptor activation enhances the therapeutic properties of opioids while attenuating unwanted side-effects such as reward and dependence that occur with sustained opioid treatment. &lt;br&gt;

Despite the recognition of both these features as part of an overreaching spectrum in any patient with AE, seldom are they the sole presenting manifestations. Additionally, the challenge is further compounded in a patient who has seronegative AE since neither sleep disturbances nor stereotypies have been well characterized with this condition yet, and the diagnosis is conditional to exhausting a list of ancillary supportive features. Fingolimod reduced the CD56bright NK cell subset. The remaining CD56dim NK cells displayed NKG2D, NKp46, CD107a, and IFN-γ levels similar to those from the patients under interferon-β1 therapy. Alternatively, comparative transcriptomics and pathway analyses revealed significant distinctions between two therapy modalities. Molecular signature of the CD56dim NK cells from fingolimod-treated MS patients was closely associated to those from healthy subjects.
