      <description>&lt;p&gt;Results In our study samples, nearly 55.8% of the older adults were found to be suffering from DS. We found a significant association between DS and being female (aOR 1.25, 95% CI 0.89-2.09), Buddhism (aOR 1.95, 95% CI 1.58-2.42), Dalits (aOR 2.60, 95% CI 1.19-5.65), unemployed, low family income (aOR 1.77, 95% CI 1.07-2.92), smokers (aOR 1.49, 95% CI 1.01-2.20) and having chronic multi-morbid conditions (aOR 1.67, 95% CI 1.09-2.55). Conclusions The prevalence of DS was high among community-dwelling older adults in eastern Nepal. Our findings suggest the need for mental health prevention and management programs targeting the older population in rural Nepal.Background Problematic internet use (PIU) is an increasingly worrisome issue, as youth population studies are establishing links with internalizing and externalizing problems. There is a need for a better understanding of psychiatric diagnostic profiles associated with this issue, as well as its unique contributions to impairment. Here, we leveraged the ongoing, large-scale Child Mind Institute Healthy Brain Network, a transdiagnostic self-referred, community sample of children and adolescents (ages 5-21), to examine the associations between PIU and psychopathology, general impairment, physical health and sleep disturbances. Methods A total sample of 564 (190 female) participants between the ages of 7-15 (mean = 10.80, SD = 2.16), along with their parents/guardians, completed diagnostic interviews with clinicians, answered a wide range of self-report (SR) and parent-report (PR) questionnaires, including the Internet Addiction Test (IAT) and underwent physical testing as part of the Healthy Brain Network protocol. Results PIU was positively associated with depressive disorders (SR aOR = 2.43, CI 1.22-4.74, p = .01; PR aOR = 2.56, CI 1.31-5.05, p = .01), the combined presentation of ADHD (SR aOR = 1.91, CI 1.14-3.22, p = .01; PR n.s.), Autism Spectrum Disorder (SR n.s.; PR aOR = 2.24, CI 1.34-3.73, p less then .001), greater levels of impairment (SR Standardized Beta = 4.63, CI 3.06-6.20, p less then .001; PR Standardized Beta = 5.05, CI 3.67-6.42, p less then .001) and increased sleep disturbances (SR Standardized Beta = 3.15, CI 0.71-5.59, p = .01; PR Standardized Beta = 3.55, CI 1.34-5.75, p less then .001), even when accounting for demographic covariates and psychiatric comorbidity. Conclusions The association between PIU and psychopathology, as well as its impact on impairment and sleep disturbances, highlight the urgent need to gain an understanding of mechanisms in order to inform public health recommendations on internet use in U.S. youth.Background Recently, it has become possible to collect next-generation DNA sequencing data sets that are composed of multiple samples from multiple biological units where each of these samples may be from a single cell or bulk tissue. Conclusions The DNA sequencing data generated by our simulator is representative of real data and integrates seamlessly with standard downstream analysis tools.Background The number of porcine Single Nucleotide Polymorphisms (SNPs) used in genetic association studies is very large, suitable for statistical testing. However, in breed classification problem, one needs to have a much smaller porcine-classifying SNPs (PCSNPs) set that could accurately classify pigs into different breeds. This study attempted to find such PCSNPs by using several combinations of feature selection and classification methods. We experimented with different combinations of feature selection methods including information gain, conventional as well as modified genetic algorithms, and our developed frequency feature selection method in combination with a common classification method, Support Vector Machine, to evaluate the method's performance. Experiments were conducted on a comprehensive data set containing SNPs from native pigs from America, Europe, Africa, and Asia including Chinese breeds, Vietnamese breeds, and hybrid breeds from Thailand. Results The best combination of feature selection methods-information gain, modified genetic algorithm, and frequency feature selection hybrid-was able to reduce the number of possible PCSNPs to only 1.62% (164 PCSNPs) of the total number of SNPs (10,210 SNPs) while maintaining a high classification accuracy (95.12%). Moreover, the near-identical performance of this PCSNPs set to those of bigger data sets as well as even the entire data set. Moreover, most PCSNPs were well-matched to a set of 94 genes in the PANTHER pathway, conforming to a suggestion by the Porcine Genomic Sequencing Initiative. However, it remains unclear how these enzymes recognize various fatty acids. Successful purification of bacterial lysophosphatidic acid acyltransferases (PlsCs) was recently reported and has paved a path for the detailed analysis of their reaction mechanisms. Here, we purified and characterized PlsC from the thermophilic bacterium Thermus thermophilus HB8. This integral membrane protein remained active even after solubilization and purification and showed reactivity toward saturated, unsaturated, and methyl-branched fatty acids, although branched-chain acyl groups are the major constituent of phospholipids of this bacterium.       <description>&lt;p&gt;Environmental complexity leads to differences in the spatial distribution of heavy metal pollution in soil and rice. Such spatial differences will seriously affect the safety of planted rice and can impact regional management and control. How to scientifically reveal these spatial differences is an urgent problem. In this study, the spatial mismatch relationship between Cd pollution in soil and rice grains (brown rice) was first explored by the interpolation method. To further reveal the causes of these, the specific recognition rules of the spatial relationship of Cd pollution were extracted based on a decision tree model, and the results were mapped. The results revealed a spatial mismatch in Cd pollution between the soil and rice grains in the study area, and the main results are as follows (i) slight soil pollution and safe rice accounted for 68.88% of the area; (ii) slight soil pollution and serious rice pollution accounted for 13.39% of the area and (iii) safe soil and serious rice pollution accounted for 11.63% of the area. In addition, 11 recognition rules of Cd spatial pollution relationship between soil and rice were proposed, and the main environmental factors were determined SOM (soil organic matter), Dis-residence (distance from residential area), soil pH and LAI (leaf area index). The average accuracy of rule recognition was 75.90%. The study reveals the spatial mismatch of heavy metal pollution in soil and crops, providing decision-making references for the spatial accurate identification and targeted prevention of heavy metal pollution spaces.Glyphosate-based herbicides (GBHs) are the most widely used pesticides for weed control. Transmembrane Transporters inhibitor In parallel with the renewal of the active ingredient, polyethoxylated POE(15) containing GBHs were banned in the EU in 2016. Since then, co-formulants were changed and numerous GBHs are marketed with different excipients declared as inert substances. In our study, we focused to determine acute and chronic cytotoxicity (by Aliivibrio fischeri assay) and direct hormonal activity (estrogenic and androgenic effects measured by Saccharomyces cerevisiae BLYES/BLYAS strains, respectively) of glyphosate, AMPA, polyethoxylated POE(15) and 13 GBHs from which 11 formulations do not contain polyethoxylated POE(15). Among the pure substances, neither glyphosate nor AMPA had any effects, while polyethoxylated POE(15) exhibited pronounced toxicity and was also estrogenic but not androgenic. Regarding the acute and chronic cytotoxicity and hormonal activity of GBHs, dilution percentages calculated from EC50 values were in the most cases by one or two order of magnitude lower than the minimum recommended dilution for agricultural and household use. Relation could not be observed between the biological effects and type of glyphosate-salts; hence toxicity could be linked to the co-formulants, which are not even declared in 3 GBHs. Toxicological evaluation must focus on these substances and free accessibility of GBHs should be reconsidered.In this study, sludge-derived biochar was prepared and utilized to support nano-zero-valent iron (NZVI-SDBC) for removing Cr(VI) and Cr(III) from aqueous solution with the aim of investigating their removal and transformation. Under the conditions of initial pH of 4, dosage of 1 g/L, temperature of 25 °C, and rotational speed of 160 rpm, 64.13% Cr species could be removed by NZVI-SDBC from Cr(VI) solution and 28.89% from Cr(III) solution. Coexisting ions experiments showed that Cu(II) and humic acids dramatically affected the removal of Cr(VI) and Cr(III), while the effect of Na(I) and Ca(II) was almost negligible. Based on this, through the coexistence and pre-loaded Cr(III) experiments, the conversion from Cr(VI) to Cr(III) was demonstrated to enhance the further attraction on Cr(VI) and promote the subsequent removal of Cr(VI). The SDBC of NZVI-SDBC could serve as electron shuttle mediator to facilitate the electron transfer between adsorbed Cr(VI) and NZVI for ortho-reduction. The transformation and removal mechanisms were further discussed by various characterizations. The kinetics of Cr(VI) removal suggested that the removal process of Cr(VI) could be divided into three phases dominated by different mechanisms (adsorption, direct/ortho reduction, electrostatic attraction), in which Cr(VI) and Cr(III) showed different behaviors of interaction. The removal of Cr(III) mainly depended on sufficient adsorption sites and the direct complexation with Fe(II). Finally, the reusability of NZVI-SDBC was assessed by adsorption/desorption recycling test. These results provided new insights into the removal and transformation mechanisms of Cr(VI) and Cr(III) by biochar-based nanocomposites.An indoor formaldehyde enriched environment was created by an automatic fumigation system with timing and concentration control. Selected hydroponic plant species were exposed in formaldehyde concentrations of 10 mg m-3, 50 mg m-3 and 100 mg m-3 respectively for 6 days with 10-h-treatment each day. Changes in morphological characteristics including leaf damage rate, leaf damage time and survival rate were monitored to evaluate morphological resistance to formaldehyde. Assessed physiological parameters were leaf chlorophyll content (Chl), leaf malondialdehyde content (MDA), activity of leaf formaldehyde dehydrogenase (FADH), leaf water soluble sugar content (WSS), and leaf proline content (Pro). Under formaldehyde suppression, reduction of Chl and increase of MDA and Pro were observed. Varying by species, FADH and WSS peaked at certain formaldehyde concentrations. A Principal Component Analysis (PCA) method was adopted to evaluate key factors in hydroponic plants' tolerance to formaldehyde. Among the 15 species selected, the best 5 performing species are Spathiphyllum floribundum, Alocasia cucullata, Davallia bullata, Syngonium podophyllum 'Pixie', and Schefflera octophylla. The study helps people to select the best ornamental plants for indoor air pollution control. The response of hydroponic plant species to formaldehyde was studied for eco-friendly indoor air pollution control.Among the most used non-steroidal anti-inflammatory drugs (NSAIDs), ketoprofen (KTF) assumes an important position. Nevertheless, its ecotoxicological effects in non-target organisms are poorly characterized, despite its use and frequency of occurrence in aquatic matrices. Thus, the aim of this study was to evaluate the possible toxicological effects of KTF contamination, in two freshwater species, Lemna minor and Daphnia magna, by measuring biochemical, physiological and population parameters. To attain this objective, both species were exposed to KTF at the same concentrations (0, 0.24, 1.2, 6 and 30 μg/L). L. minor plants were exposed during 4 d to these levels of KTF, and the enzymatic activity (catalase (CAT), glutathione S-transferases (GSTs) and carbonic anhydrase (CA)), and pigments content (chlorophylls a, b and total and carotenoids) were analyzed to evaluate the toxicity of this drug.       <description>&lt;p&gt;Regarding pregnancy-related VTE risk scores, there were 7520 patients in the low-risk group with a prenatal score less then 3 points and 239 in the high-risk group with a score ≥ 3, and 44 patients received drug prevention and treatment during pregnancy. There were 4223 patients in the low-risk group with a postpartum score less then 2 points and 3536 in the high-risk group with a score ≥ 2 points, and 824 patients received drug prevention and treatment for 10 d. Conclusion VTE poses a serious threat to maternal safety, and the society should increase its vigilance against pregnancy-related VTE.Background Postoperative unobstructed drainage is an important measure for avoiding hematoma formation and preventing complications from anterior cervical surgery. Aim To discuss the characteristics and key points of clinical management of two types of commonly used negative pressure drainage systems in clinical settings. Methods Two types of commonly used silica gel negative pressure drainage balls and a type of gastrointestinal decompression apparatus were fully emptied and then injected with different amounts of water and air. Following this, the negative pressure values of the three devices were measured. Meanwhile, we undertook a retrospective analysis of the clinical data of 1328 patients who had been treated with different negative pressure drainage apparatuses during their anterior cervical surgery in our department between January 2007 and January 2018. Results As the amount of injected air or water increased, the negative pressure of the silica gel negative pressure drainage ball decreased rapidly, dropping to zero when 150 mL of water or air was injected. In contrast, the negative pressure of gastrointestinal decompression apparatus decreased slowly, maintaining an ideal value even when 300 mL of water or air was injected. And statistical analysis demonstrated that patients who had been treated with the gastrointestinal decompression apparatus were less likely to develop severe complications than those who had been treated with the silica gel negative pressure drainage ball (P less then 0.05). Conclusion This study showed that the gastrointestinal decompression apparatus has the advantages of large suction capacity, long duration of continuous negative pressure, and good drainage effect, all of which are the favorable factors for the use of this apparatus for negative pressure drainage in anterior cervical surgery.Background Claudin 7 is often abnormally expressed in cancers and promotes the progression of some malignancies. However, the role of claudin 7 in stage II colorectal cancer (CRC) has not been studied. Aim To assess the expression and prognostic value of claudin 7 in stage II CRC. Methods We retrospectively studied 231 stage II CRC patients who underwent radical surgery at our hospital from 2013 to 2014. The protein expression level of claudin 7 was assessed and its relationship with clinicopathological features and prognosis was statistically analyzed. The independent prognostic factors were identified by Cox proportional hazards models. A prognostic grading system was constructed to stratify the survival of CRC patients. Results The expression of claudin 7 was significantly reduced in cancer tissues compared with normal tissues (P less then 0.001), and its low expression was closely related to recurrence of the disease (P = 0.017). Multivariate analysis confirmed that claudin 7 low expression (claudin 7-low) (P = 0.028) and perineural invasion positivity (PNI+) (P = 0.026) were independent predictors of poor disease-free survival (DFS). A prognostic grading system based on the status of claudin 7 and PNI classified the patients into three prognostic grades grade A (claudin 7-high and PNI-), grade B (claudin 7-low and PNI-, claudin 7-high and PNI+), and grade C (claudin 7-low and PNI+). The DFS was significantly different among the three grades (grade B vs grade A, P = 0.032; grade C vs grade A, P less then 0.001; grade C vs grade B, P = 0.040). Conclusion Claudin 7 can be used as a new prognostic marker to predict the DFS of patients with stage II CRC. The prognostic grading system with the addition of claudin 7 can further improve prognosis stratification of patients.Background Bone transport and distraction osteogenesis has been widely used to treat bone defects after traumatic surgery, but, skin and soft tissue incarceration can be as high as 27.6%. Aim To investigate the efficacy of inserting a tissue expander to prevent soft tissue incarceration. Methods Between January 2016 and December 2018, 12 patients underwent implantation of a tissue expander in the subcutaneous layer in the vicinity of a tibial defect to maintain the soft tissue in position. A certain amount of normal saline was injected into the tissue expander during surgery and was then gradually extracted to shrink the expander during the course of transport distraction osteogenesis. The tissue expander was removed when the two ends of the tibial defect were close enough. Results In all 12 patients, the expanders remained intact in the subcutaneous layer of the bone defect area during the course of transport distraction osteogenesis. When bone transport was adequate, the expander was removed and the bone transport process was completed. During the whole process, there was no incarceration of skin and soft tissue in the bone defect area. Complications occurred in one patient, who experienced poor wound healing. Conclusion The pre-filled expander technique can effectively avoid soft tissue incarceration. The authors' primary success with this method indicates that it may be a valuable tool in the management of incarcerated soft tissue.Background The Helicobacter pylori (H. pylori) infection rate in China is approximately 50%. Most patients had hematological malignancies (73%) and a low HCT-CI score (72%). The non-relapse mortality and survival were predicted according to the score. Conclusions This is one of the few studies to evaluate the HCT-CI in adults with HLA-matched donors in a developing country and our findings suggest that the high percentage of patients with a low HCT-CI scores, contrary to international reports, could be explained by different comorbidities and demographics, but mainly due to stricter filters applied to HSCT candidates and consequently, a potential selection bias caused by limited resources.Background Liquid biopsy is a novel approach for cancer diagnosis, the value of which in human gastrointestinal (GI) cancer has been confirmed by the previous studies. This article summarized the recent advances in liquid biopsy with a focus on novel technologies and the use of it in the screening, monitoring, and treatment of human GI cancer. Content The concept of liquid biopsy was first used to define the detection of circulating tumor cells (CTCs) in cancer patients, and has been expanded to other biomarkers in blood and body fluids, such as circulating tumor DNA (ctDNA), extracellular vesicles (EVs) and circulating tumor RNA. If analyzed with proper and advanced techniques like next generation sequencing (NGS) or proteomics, liquid biopsies can open an enormous array of potential biomarkers. The amount changes of target biomarkers and the mutation of genetic materials provide quantitative and qualitative information, which can be utilized clinically for cancer diagnosis and disease monitoring. Summary As a highly efficient, minimally invasive, and cost-effective approach to diagnose and evaluate prognosis of GI cancer, liquid biopsy has lots of advantages over traditional biopsy and is promising in future clinical utility. If the challenges are overcome in the near future, liquid biopsy will become a widely available and dependable option.The current climate is one of uncertainty and immeasurable tragedy for people afflicted by the pandemic of SARS-CoV-2 virus infection. As professionals, we have a duty of care towards all patients especially the vulnerable and those suffering with life-threatening illnesses such as oral cancer. Knowing factors influencing muscle thickness of the LM will provide important clues about lumbopelvic stability. selleckchem Objectives Although there are several studies in the literature investigating the adverse effects of foot-ankle postural disorders on the lumbopelvic region, to our knowledge there has been no investigation of plantar pressure distribution (PPD) as a factor influencing muscle thickness of the LM. The aim of this study was to determine whether PPD could affect LM muscle thickness. Methods This observational study consisted of 25 asymptomatic individuals. Ultrasonographic imaging was used to determine the thickness of the LM. All participants were subjected to PPD analysis using the Digital Biometry Scanning System and Milletrix software in 9 different plantar pressure zones. The Pearson product-moment correlation coefficients were used to examine the correlations between the LM muscle thickness and other variables. Stepwise multiple linear regression analysis was used to determine the variables with the greatest influence on LM muscle thickness. Results Peak pressures of medial and lateral zones of the heel were the significant and independent factors influencing static LM thickness, with 39.5% of the variance; moreover, the peak pressures of heel medial and fourth metatarsal bone were the significant and independent factors influencing dynamic LM thickness, with 38.7% of the variance. Conclusions Plantar pressure distribution could be an important factor influencing LM thickness, although further research is required. We conducted a systematic literature review of cancer genetic susceptibility studies that used NGS technologies at an exome/genome-wide scale to obtain a fuller understanding of the research landscape to date and to inform future studies. The variability across studies on methodologies and reporting was considerable. Most studies sequenced few high-risk (mainly European) families, used a candidate analysis approach, and identified potential cancer-related germline variants or genes in a small fraction of the sequenced cancer cases. This review highlights the importance of establishing consensus on standards for the application and reporting of variants filtering strategies. It also describes the progress in the identification of cancer-related germline variation to date. These findings point to the untapped potential in conducting studies with appropriately sized and racially diverse families and populations, combining results across studies and expanding beyond a candidate analysis approach to advance the discovery of genetic variation that accounts for the unexplained cancer heritability.Mechanism-based strategies to overcome resistance to programmed cell death-1 (PD-1) blockade therapy are urgently needed. We developed genetic acquired resistant models of JAK1, JAK2 and B2M loss of function mutations by gene knockout in human and murine cell lines. Human melanoma cell lines with JAK1/2 knockout became insensitive to interferon (IFN)-induced antitumor effects, while B2M knockout were no longer recognized by antigen-specific T cells and hence resistant to cytotoxicity. All of these mutations led to resistance to anti-PD-1 therapy in vivo. Objectives Prior to ratification of a local CW recommendation to limit surveillance imaging in lymphoma, we aimed to assess (A) performance characteristics of surveillance FDG-PET/CT; (B) rates, clinical consequences and costs of false positives (FP); and (C) patients and professionals' attitudes towards overuse. Methods Mixed methods (quantitative and qualitative) study. We analysed surveillance FDG-PET/CT results of two patient cohorts (n1=215 Hodgkin lymphoma and non-Hodgkin lymphoma; n2=203 Hodgkin lymphoma only). FPs were defined by negative biopsy or clinical follow-up. We held focus group discussions and in-depth interviews eliciting attce imaging, supplemented by qualitative data on patient and physician attitudes.Rationale Exposure to air pollution is linked with increased asthma morbidity and mortality. To understand pathological processes linking air pollution and allergen exposures to asthma pathophysiology, we investigated the effect of coexposure to diesel exhaust (DE) and aeroallergen on immune regulatory proteins in human airways. Methods Fourteen allergen-sensitised participants completed this randomised, double-blinded, cross-over, controlled exposure study. Each participant underwent four exposures (allergen-alone exposure, DE and allergen coexposure, particle-depleted DE (PDDE) and allergen coexposure, and sham exposure) on different order-randomised dates, each separated by a 4-week washout. Serum and bronchoalveolar lavage (BAL) were assayed for pattern recognition molecules, cytokines, chemokines and inflammatory mediators. Results In human airways, allergen-alone exposure led to accumulation of surfactant protein D (SPD; p=0.02). Coexposure to allergen and DE did not elicit the same increase of SPD as did allergen alone; diesel particulate reduction restored allergen-induced SPD accumulation. Soluble receptor for advanced glycation end products was higher with particle reduction than without it. In the systemic circulation, there was a transient increase in SPD and club cell protein 16 (CC16) 4 hours after allergen alone. CC16 was augmented by PDDE, but not DE. % eosinophils in BAL (p less then 0.005), eotaxin-3 (p less then 0.0001), interleukin 5 (IL-5; p less then 0.0001) and thymus and activation regulated chemokine (p=0.0001) were each increased in BAL by allergen. IL-5, SPD and % eosinophils in BAL were correlated with decreased FEV1. Conclusion Short-term coexposure to aeroallergen and DE alters immune regulatory proteins in lungs; surfactant levels are dependent on particle depletion.
