From preclinical ageing research to human evidence

Preclinical ageing studies can test mechanisms and generate hypotheses that would be difficult to investigate initially in people. Their value does not mean that an effect in a model already establishes a human benefit.

Preserve the model in the headline
The NIA Interventions Testing Program evaluates potential interventions in mice across multiple research sites. Its design includes reproducibility considerations and outcomes related to lifespan and later-life pathology. These remain findings in the tested model, even when they motivate further work.

When reading a report, keep the species, genetic background, sex, age at study entry and experimental setting alongside the result. A headline that omits the model can make an early-stage finding sound like completed human evidence.

Match the endpoint, not only the intervention name
Imagine a fictional cell study reporting a change in a molecular marker, followed by a mouse study reporting a functional difference. These are different outcomes at different levels. They may contribute to an evidence chain, but one does not automatically validate every interpretation of the other.

A study of survival also differs from one of a selected biomarker. Read whether the reported endpoint was planned, how long observation continued and whether adverse findings were recorded. A favourable result on one measure is not a complete account of benefit and harm.

Replication answers a specific question
Repeating a model experiment at several sites can strengthen confidence that the result is not unique to one setting. It does not by itself establish transfer to another species. Reproducibility within a model and applicability to people are separate questions.

For a comparison record, identify what stayed the same across replications and what varied. A repeated finding under one tightly controlled set of conditions may be robust there while leaving important questions about other contexts unresolved.

Recognise the missing human evidence
Human evaluation would need an appropriate population, clearly defined outcomes, safety assessment and a design suited to the proposed use. A retail product with a similar name is not automatically the material tested in a preclinical experiment, and the experiment does not authorise self-experimentation.

A responsible research summary can describe why a result is scientifically interesting, what it demonstrates in its model and what remains unknown. It need not imply delayed ageing, longer life or clinical effectiveness in people to communicate the study's contribution accurately.
