CONCLUSIONS MRI contributes to identifying PNVGB detected or suspected by ultrasonography.BACKGROUND Ischemic stroke can have a tremendously negative impact on the fitness and well-being of a child. Because endovascular interventions may be of benefit in the adult stroke population, many investigators have recently evaluated the safety and benefits of pharmacological and non-pharmacological options in the pediatric stroke population and compared pediatric patients to their adult counterparts. Some of these trials have had promising results, showing the positive effects of endovascular treatment in children with arterial acute ischemic stroke due to large-vessel occlusion (LVO). The 2015 American Heart Association/American Stroke Association guidelines suggest that mechanical thrombectomy with stent retrievers may be a consideration in some patients who are younger than 18 years and have severe LVO, when treatment (groin puncture) is initiated within 6 h of symptom onset. However, the method remains under-studied in this age group. CASE REPORT A 7-year-old girl presented with migraine-like headache, right arm and facial weakness, and expressive aphasia 9.5 h after symptom onset. Her PEDS-NIH stroke scale score was 4. Upon further investigations, she was found to have a left middle cerebral artery cryptogenic stroke with a distal left M1 clot, which was successfully treated with mechanical thrombectomy. Huge improvement was noticed during her subsequent physical examination. CONCLUSIONS Endovascular therapy offers an exciting treatment option for the management of pediatric stroke. The extent of safety of mechanical thrombectomy among children who present with large-vessel occlusion over an extended time window remains unknown and warrants further investigations.BACKGROUND Enterovirus 71 (EV71) is the pathogen most likely to cause HFMD in young children (1-5 years old). A small number of virion protein (VP) vaccine candidates are considered as the protective molecules in EV71 models. This study aimed to observe comprehensive immunogenicity for a promising EV71 vaccine depending on VP1 in neonatal mouse EV71 models. MATERIAL AND METHODS VP1 was isolated from patients and associated peptides were synthesized. EV71 particles were inactivated and mixed with Freund's complete adjuvant to prepare peptide vaccines. An EV71 vaccine was administered to establish the mouse model and the mice were infected with EV71. Hematoxylin and eosin staining was used to examine inflammatory response in EV71-infected neonatal mice. A semi-quantitative reverse transcription-polymerase chain reaction assay was performed to evaluate the levels of EV71 virus in skeletal muscle, small intestines, and brain tissues. RESULTS Three peptides were selected from 20 VP1 peptides due to their exhibition of the highest immunogenicity. The peptide injection improved inflammation and decreased EV71 particle levels in muscle, small intestines, and brain tissues. The injection also decreased lesions in the small intestines of EV71-infected mice and protected brain tissues from the EV71 infection. CX5461 CONCLUSIONS The present study confirmed the immuno-protective effects of VP1 vaccine transplantation in mice infected with EV71 virus. Our results provide valuable information that can be used in further studies investigating the specific mechanism of the anti-EV71 vaccine.The environment is becoming more and more polluted [...].The objective of this study was to develop an integrative approach in ecotoxicology (from biomarkers to population genetics) to assess the ecological status of fish populations. Flounders (Platichthys flesus) collected after the spawning season in the heavily polluted Seine estuary were compared with the moderately polluted Bay of Douarnenez. The muscle energetic reserves were highly depleted in Seine vs. Douarnenez fish. The Seine fish displaying a reduced capacity to manage the oxidative stress and a higher energetic metabolism. An increase in the content of muscle membrane phospholipids (sphingomyelin, phosphatidylserine, free sterols) was detected in the Seine vs. Douarnenez fish. The data integration allowed to hypothesize relationships between membrane phospholipids, xenobiotic metabolism, bioenergetics, and antioxidant defence. The genetic diversity considering neutral markers was maintained in the heavily polluted Seine population compared with the Douarnenez population. Finally, we suggest that the high physiological cost of tolerance to toxicants in the Seine flounder population could compromise its capacity to respond in the future to an additional stressor like warming waters in shallow depth. Thus, this population could be submitted to an ecological risk.Prodigiosins, which are natural tripyrrole red pigments and synthetic derivatives, reportedly have multiple biological effects mainly on various types of cancer cells. However, the effects of bacterial prodigiosin on non-cancerous HaCaT human skin keratinocytes have not been reported. Therefore, the present study aimed to investigate the functional activities of prodigiosin derived from cultures of the bacterium Hahella chejuensis in HaCaT cells. Cell viability, the cell proliferation rate, and reactive oxygen species (ROS) production in vitro were assayed following treatment of HaCaT cells with prodigiosin. Prodigiosin did not cause cytotoxicity and notably increased proliferation of HaCaT cells. Furthermore, prodigiosin reduced ultraviolet (UV) irradiation-induced ROS production and the inflammatory response in HaCaT cells. More importantly, prodigiosin reduced matrix metalloproteinase-9 expression and increased collagen synthesis in UV-irradiated HaCaT cells, demonstrating that it elicits anti-aging effects. In conclusion, our results reveal that H. chejuensis-derived prodigiosin is a potential natural product to develop functional cosmetic ingredients.This study aimed to investigate the neuroprotective effects of 1-methoxylespeflorin G11 (MLG), a pterocarpan, against glutamate-induced neurotoxicity in neuronal HT22 hippocampal cells. The protective effects of MLG were evaluated using MTT assay and microscopic analysis. The extent of apoptosis was studied using flow cytometric analysis performed on the damaged cells probed with annexin V/propidium iodide. Moreover, mitochondrial reactive oxygen species (ROS) were assessed using flow cytometry through MitoSOXTM Red staining. To determine mitochondrial membrane potential, staining with tetramethylrhodamine and JC-1 was performed followed by flow cytometry. The results demonstrated that MLG attenuates glutamate-induced apoptosis in HT22 cells by inhibiting intracellular ROS generation and mitochondrial dysfunction. Additionally, MLG prevented glutamate-induced apoptotic pathway in HT22 cells through upregulation of Bcl-2 and downregulation of cleaved PARP-1, AIF, and phosphorylated MAPK cascades.
